摘要
目的:观察龙膏对小鼠顺铂所致肾损伤的影响。方法:40只小鼠随机分为空白对照组、模型对照组和龙膏高、中、低剂量组,每组8只,空白对照组不作特殊处理,模型对照组及龙膏高、中、低剂量组以顺铂4 mg/kg单次腹腔注射制作肾损伤模型,龙膏高、中、低剂量组分别在给顺铂的同时予20 mg/kg/d、10 mg/kg/d、5 mg/kg/d的龙膏灌胃,模型对照组给予同体积生理盐水灌胃,给药2周后取小鼠肾脏及血清。通过肾脏HE染色切片观察其肾小管和肾小管上皮细胞形态改变。ELISA检测血清尿素氮(BUN)、肌酐(Scr)及血清胱抑素C(CysC)的含量。结果:龙膏各治疗组与模型对照组比较血清尿素氮(BUN)、肌酐(SCr)及血清胱抑素C(CysC)均下降,且中剂量龙膏治疗组下降的最明显,疗效最好,差异具有统计学意义(P<0.05)。肾脏的HE染色的病理学观察:龙膏能减轻顺铂所致肾小管的萎缩及其上皮细胞颗粒变性。结论:龙膏对小鼠顺铂所致肾损伤具有改善作用。
Objective Discussion Long gao influence on mice after cis-platinum chemotherapy,renal coefficients,blood urea nitrogen(BUN),serum creatinine(Scr),cystatin C(CysC)and renal pathology slices.Methods Cis-platinum 4 mg/kg a single intraperitoneal injection of mice to produced renal impairment,administration 5d.The mice were divided into five groups,Blank control group:Mouse feeder under conventional conditions,without any treatment;Model control group:Given the same volume of normal saline(0.2 mL/10 g)gavage;Long gao high dose group:Giving cisplatin administered simultaneously 20 mg/kg/d of high-dose Long gao gavage;Long gao middle dose group:Giving cisplatin administered simultaneously 10mg/kg/d of middle-dose Long gao gavage;Long gao low dose group:Giving cisplatin administered simultaneously 5mg/kg/d of low-dose Long gao gavage;Mice were sacrificed on day 14,take blood and renal,Using kit of ELISA detection of serum urea nitrogen(BUN)、serum creatinine(Scr)、cystatin C(CysC),renal done pathological section.Results Cisplatin after chemotherapy mice serum urea nitrogen(BUN)、serum creatinine(Scr)、cystatin C(CysC)with before chemotherapy were significantly higher,Long gao treatment serum urea nitrogen(BUN)、serum creatinine(Scr)、cystatin C(CysC)compared with Model control group decreased significantly,and middle dose Long gao treatment group decreased most significantly,the best effect.Compare with model control group,the difference was statistically significant(P<0.05).Observation of pathological changes of renal use HE staining:Blank control group:normal renal tissue structures,clear boundaries between the various structures.Model control group:multifocal renal tubular atrophy,with renal tubular compensatory hypertrophy.Tubular epithelial cell denaturation for large number of particles,diffuse infiltration of lymphocytes and monocytes,can seen a lot of tubular protein casts in renal.Three Long gao treatment groups:focal renal tubular epithelial cells granular degeneration,focal lymphocytic infiltration visible focal tubular protein casts.Conclusion Long gao can significantly reduce serum urea nitrogen(BUN)、serum creatinine(Scr)、cystatin C(CysC)content,treatment early renal damage induced by cisplatin.To change on the renal pathology is not particularly obvious,but there are still some improvement.
