犬源复合益生菌对脾虚犬抗氧化及消化吸收功能的影响 被引量:1

Effects of Canine-Derived Probiotics on Anti-oxidant,Digestive and Absorptive Functions of Splenic Asthenia Canines

摘要 试验旨在研究犬源复合益生菌对脾虚犬血清抗氧化、消化吸收功能的影响。选用24只1岁龄健康比格犬,随机分为对照组、脾虚组、自然恢复组和益生菌治疗组。对照组饲喂基础饲粮,其余3组每日饲喂基础饲粮+10 mL/kg番泻叶水煎液,灌服造模7 d。第7天,随机选取4只脾虚组犬解剖采样;对照组和自然恢复组饲喂基础饲粮;益生菌治疗组每日饲喂基础饲粮+1×10~9CFU复合益生菌制剂,持续7 d。第14天,试验犬全部解剖取样。番泻叶水煎液灌喂第7天,试验犬出现脾虚证候群,表明脾虚模型构建成功。与对照组相比,脾虚组TSOD、GSH-Px、D-Xylose和胰腺蛋白酶含量极显著降低(P<0.01),胰腺淀粉酶和脂肪酶显著降低(P<0.05),MDA含量极显著升高(P<0.01);且脾虚组十二指肠和空肠绒毛高度及V/C值显著下降(P<0.05)。透射电镜观察发现,与对照组相比,脾虚组小肠黏膜上皮细胞顶端微绒毛稀疏,细胞间连接复合体被破坏,胞内线粒体和粗面内质网发生肿胀,部分发生融合降解。脾虚犬经复合益生菌制剂治疗1周后,各项指标均恢复到对照组犬水平(P>0.05)。犬源复合益生菌能够使脾虚犬抗氧化功能和消化吸收功能恢复正常,对犬脾虚证有较好的治疗效果。 The objective of this study was to analyze the effects of canine-derived probiotics on serum antioxidant,digestive and absorptive functions of splenic asthenia canines.Twenty-four healthy beagle canines,aged 1-year-old on average,were randomly divided into 4 groups:control group,splenic asthenia group,natural recovery group and probiotic treatment group.The canines in the control group were fed with basal diet,and in the other three groups were daily fed with basal diet and 10 mL/kg of senna decoction for 7 days.On the 7th day,four canines in the splenic asthenia group were randomly dissected for collecting samples after euthanasia;the canines in the control group and the natural recovery group were fed with basal diet;the canines in the probiotics treatment group were daily fed with basal diet +1×10~9 CFU of probiotic preparation for 7 days.On the 14th day,all the experimental canines were dissected and sampled.(Results) On the 7th day,the splenic asthenia syndrome was observed in the experimental canines,indicating that the splenic asthenia model was successfully developed. Compared with the control group,the contents of T-SOD,GSH-Px,D-Xylose and pancreatic protease in the splenic asthenia group decreased significantly(P<0.01),and both the pancreatic amylase and lipase decreased(P<0.05),MDA increased significantly(P<0.01);besides,the height and V/C ratio of duodenum and jejunum decreased in the splenic asthenia group(P<0.05). Transmission electron microscopy showed that compared with the control group,the microvilli of the small intestinal mucosal epithelial cells in the splenic asthenia group were sparse,the intercellular junction complex was destroyed,the intracellular mitochondria and the rough endoplasmic reticulum were swollen,and some fusion degradation occurred.After treatment with compound probiotics for one week,all the indexes of splenic asthenia canines restored to the level of the canines in the control group(P>0.05).The canine-derived probiotics can restore the anti-oxidation,digestive and absorptive functions of splenic asthenia canines,and have a good therapeutic effect for treating splenic asthenia syndrome of canine.
