摘要
目的探讨脯氨酰4-羟化酶β亚基(P4HB)通过上皮间质转化(EMT)途径对肝癌辐射敏感性影响的机制。方法采用小干扰RNA技术,下调肝癌细胞内P4HB的表达,在体外观察P4HB对肝癌细胞转移表型影响,探讨其可能的分子机制,从而为揭示P4HB在肝癌细胞辐射敏感性中的分子机制奠定基础。结果与对照组细胞相比较,P4HB下调的肝癌细胞的辐射敏感性显著增加。接受了辐射照射之后,细胞存活率显著降低,细胞凋亡比例显著上升,G2/M期比例显著增加,而处于S期的细胞比例显著减少,放射增敏比上升(P<0.05)。结论抑制P4HB的表达,可增强肝癌辐射抵抗细胞的敏感性。
Objective To investigate the mechanism of P4HB affecting the radiation sensitivity of liver cancer through EMT pathway.Methods The expression of P4HB in hepatocellular carcinoma cells was down-regulated by small interfering RNA technique.The effect of P4HB on the metastatic phenotype of hepatocarcinoma cells was observed in vitro,and the possible molecular mechanisms were explored to reveal the molecular mechanism of P4HB in the radiation sensitivity of hepatocarcinoma cells.Results Compared with the control cells,the radiation sensitivity of liver cancer cells down-regulated by P4HB was significantly increased.After receiving radiation irradiation,the cell survival rate was significantly decreased,the proportion of apoptosis was significantly increased,and the proportion of G2/M phase was significantly increased,while the proportion of cells in S phase was significantly decreased,and the radio sensitization ratio was increased(P<0.05).Conclusion Inhibition of P4HB expression can enhance the sensitivity of liver cancer radiation-resistant cells.
