摘要
目的通过考察注射Triton WR-1339对小鼠血脂及炎症指标的影响,摸索脂代谢紊乱与炎症的关系。方法将C57BL/6随机分成6组,分别为正常组、4 h组、8 h组、36 h组、2T组,3T组。4 h、8 h、36 h组分别采用Triton WR-1339(300 mg/kg)单次肌肉注射,在相应时间点取血清及肝脏;2T组为间隔36 h进行第2次肌肉注射Triton WR-1339,末次注射36 h后取材;3T组为间隔36 h进行第3次肌肉注射Triton WR-1339,36 h后取材;测定各组中血清中总胆固醇(TC)、总三酯甘油(TG)、IL-1β、IL-6、MCP-1含量;肝脏进行HE染色,免疫组化检测肝脏中CD68、IL-1β蛋白的表达水平。结果与正常组比较,单次给予Triton WR-1339造模剂后4 h和8 h血清TC、TG无显著变化,36 h后以及2、3次重复造模后显著升高(P<0.05或0.01);MCP-1和IL-1β在造模8 h后血清中含量明显增加(P<0.05),多次注射后显著性升高(P<0.01);而IL-6仅在第3次注射后才明显增加(P<0.05);肝脏在造模8 h开始出现明显的炎症浸润,随注射次数炎症面积显著增加(P<0.05或0.01);肝脏IL-1β表达在造模4 h后显著增加,CD68表达在造模8 h后增加(P<0.05或0.01)。结论Triton WR-1339所致肝脏炎症反应先于高脂血症的发生,且与MCP-1及M1巨噬细胞形成有密切关系。
Objective To explore the relationship between lipid metabolism disorder and inflammation through investigating the effect of Triton WR-1339 on lipid and inflammation in mice.Methods C57BL/6 mice were randomly divided into 6 groups,including the normal group,4 h group,8 h group,36 h group,2 T group and 3T group.Mouse serum and liver were taken after Triton WR-1339(300 mg/kg)treatment by single intramuscular injection at 4 h,8 h and 36 h,respectively.In 2T group,Triton WR-1339 was injected twice at a 36 h interval.In 3 T group,Triton WR-1339 was injected three times at a 36 h interval.Serum and liver were taken after the last 36 h injection.The contents of TC,TG,IL-1β,IL-6 and MCP-1 in serum were determined.The liver was stained with H&E,and the expression levels of CD68 and IL-1βproteins in the liver were detected by immunohistochemistry.Results Compared with the normal group,there was no significant changes in serum TC and TG at 4 h and 8 h after single administration of Triton WR-1339,but it increased significantly after 36 h(P<0.05 or 0.01).The serum content of MCP-1 and IL-1βincreased at 8 h(P<0.05),which increased significantly after multiple injections(P<0.01).And the serum content of IL-6 increased after three-time injection.After 8 h of modeling,the liver began to show obvious inflammatory infiltration,and the inflammatory area increased significantly after multiple injections(P<0.05 or 0.01).And the expression of IL-1βin liver was increased significantly after 4 h of modeling,and CD68 was elevated after 8 h.Conclusion WR-1339 induces inflammation of the liver prior to hyperlipidemia,which is closely related to MCP-1 and M1 macrophage formation.
