摘要
目的研究原肌球蛋白调节蛋白1(Tmod1)对载脂蛋白E(ApoE)基因敲除小鼠的糖脂代谢的影响。方法将心脏过表达而全身其他部位敲除Tmod1的小鼠(TOT/Tmod1-/-)与ApoE-/-小鼠杂交获得ApoE和Tmod1的双敲小鼠(ApoE-/-/TOT/Tmod1-/-)。对ApoE-/-小鼠和双敲小鼠进行高脂饮食喂养,检测2种小鼠的体质量改变、脂肪含量、血脂水平、口服糖耐量(OGTT)和胰岛素耐量(ITT)。对肝脏和皮肤进行组织学分析。结果双敲小鼠较ApoE-/-小鼠体质量减轻,体脂含量降低(P<0.05)。但喂养高脂饮食8周后,双敲小鼠体质量增加量、皮下脂肪组织的厚度、三酰甘油和总胆固醇水平均高于ApoE-/-小鼠(P<0.05),而脂肪肝、口服葡萄糖耐量和胰岛素耐受水平则明显降低(P<0.05)。结论Tmod1参与调节高脂饮食引起的ApoE-/-小鼠糖脂代谢水平的改变。
Objective To study the effect of Tropomodulin1(Tmod1)on the glucose and lipid metabolisms of ApoE knockout mice.Methods Tmod1 knockout mice with heart specific Tmod1 overexpressing(TOT/Tmod1-/-)were bred with ApoE-/-to obtain ApoE and Tmod1 double knockout mice(ApoE-/-/TOT/Tmod1-/-).ApoE-/-and ApoE-/-/TOT/Tmod1-/-mice were fed with high-fat diet;the changes in body weight,body fat content and blood lipid were measured.The oral glucose tolerance test(OGTT)and insulin tolerance test(ITT)were performed.Histological analyses were performed on the liver and the skin.Results ApoE-/-/TOT/Tmod1-/-mice had less body weight and body fat than ApoE-/-mice(P<0.05).After fed with high-fat diet for 8 weeks,ApoE-/-/TOT/Tmod1-/-mice showed much greater body weight change,higher triglyceride and total cholesterol levels,thicker subcutaneous apdipose tissues than ApoE-/-mice(P<0.05).However,the fat deposit in the liver,the level of oral glucose tolerance as indicated by the level of glucose being relatively higher during the OGTT,and the rate of insulin-stimulated blood glucose disposal dring the ITT of ApoE-/-/TOT/Tmod1-/-mice were lower than those of ApoE-/-mice(P<0.05).Conclusion Tmod1 is involved in the regulation of glucose and lipid metabolisms of ApoE-/-mice induced by high-fat diet.
