短期热应激对肉仔鸡生长性能及腺苷酸活化蛋白激酶信号通路的影响 被引量:3

Effects of Short-Time Heat Stress on Growth Performance and Adenosine 5’-Monophosphate Activated Protein Kinase Signal Pathway of Broiler Chickens

摘要 本试验旨在研究短期热应激对肉仔鸡生长性能及食欲中枢腺苷酸活化蛋白激酶(AMPK)信号通路的影响。选取体重相近的28日龄雄性爱拔益加肉仔鸡72只,随机分为热应激组和对照组,每组6个重复,每个重复6只鸡。热应激组温度设定为(33±1)℃,对照组温度设定为(25±1)℃,试验持续10 h。试验结束后,统计肉仔鸡生长性能,每个重复取1只鸡采集血浆和下丘脑样品,测定血浆尿酸(UA)、葡萄糖(GLU)、甘油三酯(TG)和总胆固醇(TCHO)含量,检测下丘脑大麻素1(CB1)、丝氨酸/苏氨酸蛋白激酶1(AKT1)、AMPKα1、AMPKα2、肝激酶B1(LKB1)、哺乳动物雷帕霉素靶蛋白(mTOR)、糖皮质激素受体(GR)的mRNA相对表达量和AMPK磷酸化水平。结果表明:1)与对照组相比,热应激组肉仔鸡体增重和采食量显著降低(P<0.05)。2)与对照组相比,热应激组肉仔鸡血浆UA、GLU和TCHO含量均显著提高(P<0.05)。3)与对照组相比,热应激组肉仔鸡下丘脑中AMPKα1和G R的mRNA相对表达量显著降低(P<0.05),热应激组肉仔鸡下丘脑中AMPK磷酸化水平显著降低(P<0.05),热应激组肉仔鸡下丘脑中LKB1、AKT1、mTOR、CB1和AMPKα2的mRNA相对表达量均无显著差异(P>0.05)。由此可见,短期热应激能显著降低肉仔鸡生长性能和下丘脑中AM PK磷酸化水平,显著降低下丘脑中AMPKα1和GR的mRNA相对表达量,提示AMPK信号通路可能参与短期热应激抑制肉仔鸡食欲的调控过程。 This trial was conducted to investigate the effects of short-time heat stress on growth performance and adenosine 5’-monophosphate activated protein kinase(AMPK) signal pathway in appetite centers of broilers. Seventy-two 28-day-old Arbor Acres broilers with similar weight were randomly allocated into 2 groups(heat stress group and control group) with 6 replicates per group and 6 chicks per replicate. The ambient temperature in the heat stress group was(33±1) ℃,and the ambient temperature in the control group was(25±1) ℃,the experiment last for 10 h. At the end of the experiment,statistic growth performance of broilers,and the plasma and hypothalamus samples were collected from 1 chicken in each replicate. The contents of uric acid(UA),glucose(GLU),triglyceride(TG) and total cholesterol(TCHO) in plasma,and the mRNA relative expressions of cannabinoid 1(CB1),serine/threonine-protein kinase 1(AKT1),AMPKα1,AMPKα2,hepatokinase B1(LKB1),mammalian target of rapamycin(mTOR) and glucocorticoid receptor(G R) in hypothalamus of broilers were detected,as well as the phosphorylation level of AMPK. The results showed as follows: 1) compared with the control group,the feed intake and body weight gain of broilers in heat stress group were significantly decreased(P<0.05). 2) Compared with the control group,the contents of GLU,UA and TCHO in plasma of broilers in heat stress group were significantly increased(P<0.05). 3) Compared with the control group,the mRNA relative expressions of AMPKα1 and G R in hypothalamus of broilers in heat stress group were significantly decreased(P<0.05),the phosphorylation level of AMPK in hypothalamus of broilers in heat stress group was significantly decreased(P < 0. 05),and the mRNA relative expressions of LKB1,AKT1,mTOR,CB1 and AMPKα2 in hypothalamus of broilers in heat stress group had no significant difference(P>0.05). In conclusion,short-time heat stress can significantly decrease the growth performance and phosphorylation level of AMPK in hypothalamus of broilers,significantly decrease the mRNA relative expressions of AMPKα1 and G R in hypothalamus,and point out that AMPK signal pathway may be involved in the regulation of appetite inhibition by short-time heat stress in broilers.[Chinese Journal of Animal Nutrition,2019,31(12) : 5533-5540]
