移植同种异体脂肪干细胞对兔椎间盘退变早期的干预实验研究 被引量:2

Experimental study on the transplantation of allogeneic adipose derived stem cells for the early stage of intervertebral disc degeneration in rabbits

摘要 目的观察同种异体兔脂肪干细胞(adipose derived stem cells,ADSC)在退变早期对椎间盘的修复效果。方法采用穿刺抽吸法建立兔椎间盘退行性变模型,体外培养ADSC对模型进行移植治疗。27只5月龄日本大耳白兔随机分为3组:正常对照组(NC组,n=9),ADSC移植组(ADSC组,n=9)和DMEM培养基组(DMEM组,n=9)。每只实验动物的L 3-4、L 4-5和L 5-6作为实验干预椎间盘。术后4,8,12周分别利用标准化T2加权像信号强度(%ST2WI)、病理组织学、退变分数、免疫组化染色以及Ⅱ型胶原(collagenⅡ)和蛋白聚糖(aggrecan)mRNA表达水平评价修复效果。结果术后4,8,12周,DMEM组的%ST2WI值、collagenⅡ和aggrecan的mRNA表达量均明显低于其他各组(P<0.05),NC组和ADSC组组间无明显差异;NC组和ADSC组退变分数明显低于DMEM组(P<0.05),NC组和ADSC组间无明显差异;NC组和ADSC组的collagenⅡ免疫组化均显示强阳性,而DMEM组随着时间的推移阳性逐渐减弱。结论在退变早期移植同种异体的兔ADSC能有效抑制椎间盘的退变。 Objective To observe the effect of allogeneic rabbit adipose stem cell(ADSC)on the repair of intervertebral disc in the early stage of degeneration.Methods The intervertebral disc degeneration animal model was constructed by the conventional method and at the same time ADSC was transplanted into the model rabbits of disc degeneration.Twenty-seven 5-month-old Japanese white rabbits were randomly divided into 3 groups(n=9 in each group):normal control(NC)group,ADSC transplantation group(ADSC group)and DMEM group.L 3-4,L 4-5 and L 5-6 intervertebral discs of each rabbit were treated in the experiment.At week 4,8 and 12 after the transplantation,the repair effect of rabbit degenerative intervertebral discs was evaluated by standard T2-weighted image ratio(%ST2WI),histology,degeneration score,immunochemical staining and mRNA expression levels of collagenⅡand aggrecan.Results At week 4,8 and 12 after cell-transplantation,standard T2-weighted image ratio,mRNA expression levels of collagenⅡand aggrecan in DMEM group were significantly lower than those in the other groups(P<0.05),but no significant difference was found between NC group and ADSC group.Histological analysis indicated that the degeneration score in NC group and ADSC group was significantly lower than that in DMEM group(P<0.05),but there was no significant difference between NC group and ADSC group.Immunochemical staining of collagenⅡindicated strongly positive in NC group and ADSC group at different time,but it gradually weakened with the increase of time in DMEM group.Conclusion The results suggest that ADSC may effectively restrain the disc degeneration.
