摘要
目的建立十一味活血酊的HPLC指纹图谱,并同时测定血竭素高氯酸盐、延胡索乙素、胡椒碱三种指标成分的含量,为十一味活血酊的质量控制研究提供参考依据。方法通过高效液相色谱仪,采用Shim Pack CLC ODS色谱柱(4.6 mm×250 mm,5µm),以乙腈-0.1%磷酸溶液为流动相,梯度洗脱;检测波长:280 nm;柱温:40℃;流速:1.0 ml/min。采用“中药色谱指纹图谱相似度评价系统”建立10批十一味活血酊指纹图谱,并用HPLC法同时测定3种成分的含量。结果确定了十一味活血酊的HPLC指纹图谱共有模式,标定了41个共有峰,各峰分离度良好,相似度均在0.91以上,并同时测定了10批样品中血竭素高氯酸盐、延胡索乙素、胡椒碱三种成分的平均含量,分别为17.824、37.955、18.229 mg/g。结论建立的HPLC指纹图谱分析方法与多指标成分测定方法简便可行、稳定可靠,为十一味活血酊的质量控制研究提供科学依据。
Objective To establish the HPLC fingerprint of Shiyiwei Huoxue tincture and determine the contents of three kinds include dracorhodin perchlorate,tetrahydropalmatine and piperine of index components simultaneously to provide reference for the quality control of Shiyiwei Huoxue tincture.Methods The Shim Pack CLC ODS(4.6 mm×250 mm,5μm)is chosen with acetonitrile and 0.1%phosphoric acid solution as mobile phase.The detection wavelength is set at 280 nm,and the column temperature is set at 40℃,and the flow rate is 1.0 ml/min.The chromatographic fingerprint evaluation system published by the State Pharmacopoeia Commission is used to establish the fingerprint of 10 batches of Shiyiwei Huoxue tincture,and the contents of three kinds of components are determined simultaneously by the established HPLC method.Results The characteristic fingerprint peak of Shiyiwei Huoxue tincture is composed of 41 chromatographic peaks,and the resolution of each peak is good,and the similarity is higher than 0.91,and the average content of dracorhodin perchlorate,tetrahydropalmatine and piperine are 17.824mg/g,37.955mg/g,18.229mg/g,respectively.Conclusion The established HPLC fingerprinting analysis method and multi-index components determination method are simple and feasible to provide reference for the quality control of Shiyiwei Huoxue tincture.
