摘要
数字PCR是一种新兴的基因检测方法,是继常规PCR、实时荧光定量PCR之后的第三代PCR基因检测技术.在传统的水产病原菌的分子检测方法中,普通PCR检测结果需要通过琼脂糖凝胶电泳检测、耗时长,并且不能对检测病原进行定量;实时荧光定量PCR技术则需要依赖内参基因进行相对定量分析,难以满足水产病害快速精准检测的需要.数字PCR技术实现了对检测样本的绝对定量分析,具有前所未有的准确度和重现性,为快速准确地进行水产病原菌的检测提供了崭新的技术平台,具有广阔的发展和应用前景.本文主要介绍了数字PCR的产生、原理和分类,并深入探讨了数字PCR技术在水生生物致病菌和水产品食源性致病菌检测中的应用.
The digital PCR technology is a new method for gene detection,which is the third generation of PCR gene detection technology after conventional PCR and real-time fluorescence quantitative PCR.In the traditional molecular detection method of aquatic pathogenic bacteria,the common PCR products need to be detected by agarose gel electrophoresis,which is long time consuming and cannot detect pathogenic bacteria quantitatively.Real-time fluorescence quantitative PCR technology relies on internal reference genes for relative quantitative analysis,which is difficult to meet the needs of detecting the aquatic diseases rapidly and accurately.By contrast,digital PCR technology can achieve absolute quantitative analysis,with unprecedented accuracy and reproducibility.Digital PCR has broad application prospects,providing a new technology platform for rapid and accurate detection of aquatic pathogens.This paper mainly introduced the generation,principle and classification of digital PCR.The application of digital PCR to detect the aquatic pathogens and foodborne pathogens of aquatic products was also discussed.
