摘要
【目的】探究CsAN1在花青素合成调控中的作用。【方法】运用RT-PCR技术比较了不同茶树品种(系)中CsAN1的表达差异,利用F1作图群体和SNP标记对CsAN1进行了连锁定位,分析了该基因的表达量、基因型与新梢颜色间的相关性。【结果】①在7个供试品种的春梢第二叶中,CsAN1在紫色品种(系)中的表达量显著高于常规品种;②设计引物扩增得到441 bp的Cs AN1基因片段,直接测序发现2个SNP位点,利用它们在作图群体中的基因型数据将CsAN1定位到茶树LG08号连锁群上;③结合前期对作图群体新梢颜色的观测和QTL定位结果,发现CsAN1的不同基因型单株新梢颜色评级差异极显著(P<0.01),且该基因与控制茶树新梢颜色的一个主效QTL(qYSC8)重合。【结论】从遗传学角度进一步证实了CsAN1参与了茶树花青素合成过程的调控。
【Objective】The objective of the study was to measure that CsAN1 plays an important role in the anthocyanin synthesis in C. sinensis.【Method】We analyzed the expression levels of CsAN1 in seven different tea cultivars(lines) by RT-PCR technique,genotyped CsAN1 in a F1 mapping population with SNP markers. Furthermore,the relationships between expression levels,genotypes and young shoot color of those tea plants were analyzed.【Result】①In the second leaves of spring shoots of the seven tested cultivars,CsAN1 expression levels in the high-anthocyanin cultivars(lines) was significantly higher than that of normal cultivars. ②A 441 bp gene fragment of CsAN1 was amplified and two SNPs were found.Using the SNPs genotypes in a mapping population(n=55),CsAN1 was mapped on the LG08 linkage group of C. sinensis. ③Based on the previous results of young shoot color(YSC) observation and QTL mapping,we found that the YSC scores of the mapping population were significantly different between two genotypes of CsAN1(P<0.01),and this gene were co-localized with a major QTLs(qYSC8) that controlling the young shoot color of tea plants.【Conclusion】The results of this study further confirmed that CsAN1 plays an important role in the anthocyanin synthesis in C. sinensis.
