川芎嗪对高糖作用下人晶状体上皮细胞凋亡的影响 被引量:1

Effect of ligustrazine on apoptosis of human lens epithelial cells on high glucose condition

摘要 目的观察川芎嗪注射液对高糖作用下人晶状体上皮细胞(HLEs)凋亡的影响。方法体外培养HLE-B3,将传代的细胞随机分为正常对照组、高糖组、高糖+川芎嗪组,分别用含5.5 mmol/L葡萄糖、25 mmol/L葡萄糖、25 mmol/L葡萄糖+0.2 μg/mL川芎嗪的培养基培养48 h。TUNEL染色法检测3组HLEs的凋亡率,Western blot法检测3组促凋亡蛋白C aspase-3及抑制凋亡蛋白Bcl-2的表达。结果高糖组HLEs凋亡率高于正常对照组及高糖+川芎嗪组(P<0.05),高糖+川芎嗪组与正常对照组比较差异无统计学意义(P>0.05)。与正常对照组比较,高糖组Caspase-3表达升高(P<0.05),高糖+川芎嗪组表达无明显变化(P>0.05);与正常对照组比较,高糖组Bcl-2表达降低(P><0.05),高糖+川芎嗪组差异无统计学意义(P>0.05)。结论川芎嗪可抑制高糖作用下HLEs凋亡的发生。 Objective To observe the effect of ligustrazine injection on apoptosis of human lens epithelial cells stimulated by high glucose.Methods The HLEs(human lens epithelial cells,H LEs)were cultured in vitro and the subcultured cell were divided into three groups,namely the normal glucose group(5.5 mmol/L glucose,N),the high glucose group(25 mmol/L glucose,N),and high glucose+ligustrazine group(including 0.2μg/m L of ligustrazine medium,H+L).Apoptosis rate of lens epithelial cells in 3 groups was detected by TUNEL staining.The proteins expression levels of apoptosis-promoting protein Caspase-3 and inhibition of apoptosis protein Bcl-2 in three groups were detected by Western blot.Results The apoptosis rate of lens epithelial cells was higher in the high glucose group than in the normal control group and the ligustrazine group(P<0.05),and the difference was not statistically significant between the ligustrazine group and in the normal control gro u p(P>0.05).The expression of caspase-3 was higher in the high-glucose group than in the normal control group(P<0.05),while the expression of caspase-3 was not significantly changed in the ligustrazine group(P>0.05).The expression of Bcl-2 was down-regulated in the high glucose group(P<0.05)compared with normal control group,and the difference was not statistically significant between the ligustrazine group and the normal control group(P>0.05).Conclusion Ligustrazine can inhibitthe apoptosis of humman lens epithelial cells induced by high glucose.
