Bioinformatics analysis of DCT gene in Red fox

摘要 为了探讨赤狐DCT基因及其所编码蛋白的性质和功能,进而为挖掘赤狐毛色基因调控机理提供理论基础,试验利用生物信息学分析方法对NCBI中公布的赤狐DCT基因序列(登录号为XM_026003622.1)的编码序列(CDS区)及其编码蛋白的理化性质、跨膜结构域、亚细胞定位、磷酸化位点、信号肽、高级结构进行了分析。结果表明:赤狐DCT基因CDS区全长1650 bp,共编码549个氨基酸;编码蛋白等电点为6.07,含有20种氨基酸,其中亮氨酸(Leu)含量最高;编码蛋白为不稳定亲水性蛋白;该蛋白主要定位于细胞质、分泌小泡、线粒体等细胞器中,为跨膜蛋白;该蛋白存在43个磷酸化位点,且不存在信号肽;DCT蛋白主要为无规则卷曲结构;该基因保守性较好,且与其遗传关系最近的为家犬。 The aim of the present study was to identify DCT gene,analyze the quality and function of its encoded protein,and provide theoretical basis for exploring the regulation mechanism of hair color gene in red fox.Bioinformatics analysis was used to analyze the CDS region of red fox DCT gene sequence(login number:XM_026003622.1)published on NCBI website and the physical and chemical properties,transmembrane structural domain,subcellular localization,phosphorylation site,signal peptide and advanced structure of the encoded protein.The results showed that the CDS region of DCT gene of Red fox was 1650 bp,encoding 549 amino acids.The isoelectric point of the encoded protein was 6.07,containing 20 amino acids,of which leucine(Leu)was the highest,which was an unstable hydrophilic protein.This protein is mainly located in cytoplasm,secretory vesicles,mitochondria and other organelles,and is a transmembrane protein.The protein has 43 phosphorylation sites and no signal peptide.The advanced structure of DCT protein was mainly random coil.The DCT gene was highly conservative and the closest genetic relationship was domestic dogs.
