摘要
组蛋白甲基化修饰是当前表观遗传学研究的重要内容之一。组蛋白的甲基化由组蛋白甲基化酶和组蛋白去甲基化酶动态调节。组蛋白赖氨酸去甲基化酶5A[lysine(K)demethylase 5A,KDM5A]是组蛋白去甲基化酶家族的重要成员,它可以特异性去除组蛋白H3第4位赖氨酸上的二甲基和三甲基(H3K4me2/3),从而介导基因沉默,并调节细胞功能。KDM5A可以直接或间接地保持肿瘤细胞干性,抑制细胞代谢和分化,促进肿瘤细胞增殖、转移和耐药,因此其与多种肿瘤的发生、发展及耐药密切相关,有望成为肿瘤诊断和治疗的新的潜在靶点。
Histone methylation is one of the important research contents of epigenetics. Methylation of histones is dynamically regulated by histone methylase and histone demethylase. Histone lysine(K)demethylase 5 A(KDM5 A) is an important member in the histone demethylase family, which can specifically remove the dimethyl and trimethyl from the 4 th lysine of histone H3(H3 K4 me2/3),thus mediating gene silencing and regulating cell function. KDM5 A can directly or indirectly maintain tumor cell dryness, inhibit cell metabolism and differentiation, promote tumor cell proliferation,metastasis and drug resistance, so it is closely related to the occurrence, development and drug resistance of various tumors.KDM5 A may become a new potential target for the diagnosis and treatment of tumors.
