摘要
目的:探讨补中益气汤对肺腺癌荷瘤裸鼠肿瘤细胞A549/DDP上皮-间质转化(EMT)的影响。方法:BALB/C裸鼠随机分为空载(A)组,模型(B)组,顺铂(C)组和补中益气汤低(D)、中(E)、高(F)剂量组。A组接种TGF-β1空载慢病毒转染的A549/DDP细胞,其余组接种TGF-β1稳定过表达的A549/DDP细胞。干预后处死裸鼠,取肿瘤组织。称量瘤重,计算抑瘤率;HE染色观察肿瘤细胞形态;免疫组化、Western Blot、Real-time PCR检测EMT分子标志物波形蛋白(Vim)、N-钙黏蛋白(N-cad)、α-平滑肌肌动蛋白(α-SMA)、E-钙黏蛋白(E-cad)、α-连环蛋白(α-CA)的表达情况。结果:与B组比较,D、E、F组瘤重减小,抑瘤率上升(P<0.05);B组细胞纺锤样改变,D、E、F组细胞圆形或椭圆形增多;与B组比较,E、F组Vim、N-cad、α-SMA蛋白及mRNA表达均下调(P<0.05),E-cad、α-CA蛋白及mRNA表达均上调(P<0.05);各中药组EMT不同程度改善。结论:补中益气汤对肺腺癌A549/DDP细胞EMT有明显的抑制作用,并具有一定的剂量依赖性。
Objective:To investigate the effects of Buzhong Yiqi Decoction on epithelial-mesenchymal transition(EMT)of tumor cells A549/DDP in mice with lung adenocarcinoma.Methods:BALB/C nude mice were randomly divided into empty control group(A group),model group(B group),cisplatin group(C group),and Buzhong Yiqi Decoction groups with different doses(D,E,F group).A group was inoculated with A549/DDP cells transfected by uninfected lentiviral TGF-β1,and the other groups were inoculated with TGF-β1 over-expression A549/DDP cells.After the intervention,the nude mice were sacrificed and the tumor tissue was taken.The tumors were weighed,and inhibition rate was calculated;HE staining was used to observe cell morphology;Immunohistochemistry,Western blot and Real-time PCR were used to detect the expression of EMT molecular markers Vimentin(Vim),N-cadherin(N-cad),α-smooth muscle agonist protein(α-SMA),E-cadherin(E-cad),α-catenin(α-CA).Results:Compared with B group,the weight of tumors decreased gradually,while the inhibition rate increased in D,E,F group(P<0.05).Spindle-like changes were common in group B cells,the number of round or oval cells increased gradually in D,E,F group.Compared with B group,the expression of protein and mRNA of Vim,N-cad,α-SMA was down-regulated(P<0.05),and the expression of E-cad,α-CA was up-regulated in E,F group(P<0.05),F group was better.The EMT of each Chinese herbal medicine group was improved in varying degrees.Conclusion:Buzhong Yiqi Decoction inhibits EMT of lung adenocarcinoma A549/DDP tumor cells in a dose-dependent manner.
