基于网络药理学探讨人参茯苓药对治疗老年性痴呆的作用机制 被引量:22

Mechanism of Radix Ginseng-Poria Drug Pair on Senile Dementia Based on Network Pharmacology

摘要 目的探讨人参-茯苓药对治疗老年性痴呆的潜在生物学机制。方法利用TCMSP数据库结合文本挖掘方式,收集人参、茯苓两味药物所含有的活性成分,并进行口服利用率、血脑屏障透过率和药物相似性等参数筛选。通过化学成分靶点预测和靶点注释,利用Cytoscape 3.6软件进行活性成分靶点网络和靶点-疾病网络的构建及拓扑分析,预测重要的成分与靶点。利用DAVID数据库对人参-茯苓药对的靶点进行GO分析和KEGG富集分析。结果筛选出人参-茯苓药对28个活性成分,其中人参22个,茯苓6个;得到89个药物靶点,包括雌激素受体、雄激素受体、孕激素受体、毒蕈碱乙酰胆碱受体M3、毒蕈碱乙酰胆碱受体M1等。发现了该药对的10个潜在治疗疾病分别是神经系统疾病、内分泌系统疾病、免疫系统疾病、心血管疾病、精神障碍、代谢疾病、营养障碍、消化系统疾病、细菌感染或真菌病、癌症。富集分析与老年性痴呆相关的10个潜在信号通路包括神经活性配体-受体相互作用、钙信号通路、胆碱能突触、肾素分泌、雌激素信号通路等。结论人参-茯苓药对治疗老年性痴呆具有多靶点、多途径、多通路的特点,其主要生物学机制可能与神经活性配体-受体相互作用、钙信号通路、胆碱能突触等信号通路有关。 Objective To study the underlying biological mechanism of Radix Ginseng-Poria drug pair in the treatment of senile dementia on the basis of network pharmacology.Methods The active components of Radix Ginseng and Poria were collected by using TCMSP database and text mining,and the parameters such as oral utilization rate,blood-brain barrier permeability and drug similarity were screened.Through chemical component target prediction and target comments,the active component-target network and target-disease network were constructed and topological analysis was conducted using Cytoscape 3.6 software to predict the important components and targets.GO analysis and KEGG enrichment analysis of the target of Radix Ginseng and Poria were performed with the DAVID database.Results The 28 active ingredients of Radix Ginseng-Poria drug pair were screened,including 22 in Radix Ginseng and 6 in Poria;89 drug targets were obtained,including estrogen receptor,androgen receptor,progesterone receptor and acetylcholine receptor M3,acetylcholine receptor M1,etc.The 10 potential treatment diseases of the drug pair were found to be neurological diseases,endocrine system diseases,immune system diseases,cardiovascular diseases,mental disorders,metabolic diseases,nutritional disorders,digestive diseases,bacterial infections or fungal diseases,and cancer.Enrichment analysis of 10 potential signaling pathways associated with senile dementia included neuroactive ligand-receptor interactions,calcium signaling pathways,cholinergic synapses,renin secretion,and estrogen signaling pathways.Conclusion Radix Ginseng-Poria drug pair has the characteristics of multiple targets,multiple pathways and multiple channels in treating senile dementia.Its main biological mechanism may be related to neuroactive ligand-receptor interaction,calcium signaling pathway,and cholinergic synaptic signaling pathways.
