白虎加人参汤含药血浆对高糖环境下SD大鼠胰岛细胞增殖、凋亡及氧化应激水平的影响 被引量:14

Effects of Baihu Jia Renshen Decoction(白虎加人参汤) Containing Plasma on Islet Cell Proliferation,Apoptosis and Oxidative Stress Under High Concentration D-glucose in SD Rats

摘要 目的探讨白虎加人参汤治疗2型糖尿病的可能作用机制。方法将28只SD大鼠随机分成空白组(无菌超纯水)、白虎加人参汤组(浓度为2 g/ml)、二甲双胍组(浓度为21.5 mg/ml)、金匮肾气丸组(浓度为0.215 g/ml),每组7只,每日灌胃1次,33.48 ml/kg,连续5天,于末次灌胃后麻醉腹主动脉取血制备含药血浆。将大鼠胰岛瘤B细胞株INS-1细胞分为正常组、模型组、白虎加人参汤含药血浆组、二甲双胍含药血浆组、金匮肾气丸含药血浆组,每组3个复孔。除正常组外其余各组以高浓度葡萄糖(33.3 mmol/L)处理细胞构建细胞高糖损伤模型。正常组和模型组正常培养,其余各组分别用5%相应含药血浆培养24 h后,比较各组INS-1细胞增殖情况、细胞凋亡率,检测细胞内活性氧簇(ROS)水平和总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性,分别观察各组细胞在基础葡萄糖刺激下和高糖刺激下胰岛素释放水平。结果与正常组比较,模型组细胞增殖OD值降低,细胞凋亡率和细胞内ROS水平升高,SOD及T-AOC活性下降,在基础葡萄糖刺激和高糖刺激后胰岛素分泌水平均明显下降(P<0.01)。与模型组比较,各药物血浆组细胞增殖OD值升高,细胞凋亡率和细胞内ROS水平降低,SOD及T-AOC活性升高,在基础葡萄糖刺激和高糖刺激后胰岛素分泌水平均升高(P<0.05或P<0.01),而二甲双胍含药血浆组和白虎加人参汤含药血浆组改善较金匮肾气丸含药血浆组明显(P<0.01),白虎加人参汤含药血浆组与二甲双胍含药血浆组作用相当(P>0.05)。结论白虎加人参汤可有效改善高糖诱导引起的INS-1细胞损伤和凋亡,改善细胞胰岛素分泌功能,其作用与其提高了INS-1细胞的抗氧化能力、降低细胞内ROS水平有关。 Objective To study the possible mechanism of Baihu Jia Renshen Decoction(白虎加人参汤)treating type 2 diabets.Method Totally 28 SD rats were divided into four groups,blank group(treated with aseptic ultrapure water),Baihu Jia Renshen Decoction group(concentration 2 g/ml),metformin group(concentration 21.5 mg/ml),and Jingui Shenqi Pills group(concentration 0.215 g/ml),7 rats in each group.All rats were treated with gastric perfusion 33.48 ml/kg for continuous five days.Drug-containing plasma was prepared by taking blood sample under abdominal aorta anaesthesia after the last perfusion.Isletoman B cell strain INS-1 were divided into five groups,normal group,model group,Baihu Jia Renshen Decoction containing plasma group,metformin containing plasma group,Jingui Shenqi Pill and containing plasma group,3 wells for each group.All groups but normal group were treated with high-concentration glucose(33.3 mmol/L)to process cells and build high-glucose injury model.The normal group and model group were cultured normally,and other groups were cultured with 5%corresponding drug containing plasma for 24 h.INS-1 cell proliferation,apoptosis rate,intracellular active oxygen species(ROS)level and total antioxidant capacity(T-AOC),super-oxide dismutase(SOD)level were compared.Insulin release level of all groups under basic and high glucose stimulation was observed.Result Compared with the normal group,the proliferation activity of the model group was significantly inhibited,the apoptotic rate and ROS increased significantly,the content of SOD and T-AOC were decreased,insulin secretion under basic or high glucose stimulation decreased significantly compared with the model group,all drug-containing plasma treated group showed cell proliferation OD increase,appotosis rate decrease,ROS level decrease,SOD and T-AOC activity increase and insulin secretion level increase under basic or high glucose stimulation(P<0.05 or P<0.01).The improvement of melformin containing group and Baihu Jia Renshen Decoction containing group was better than that of the Jingui Shenqi Pill containing group,while the improvement of Baihu Jia Renshen Decoction containing group and melformin containing group was quite(P>0.05).Conclusion Baihu and Renshen Decoction markedly improve high-glucose induced INS-1 cell injury and apoptosis,improving insulin secretion,and the action might be related to increasing INS-1 cell antioxide capacity and reducing intracellular ROS level.
