摘要
通过单因素试验对实验室筛选鉴定的菌株Paraburkholderia caffeinitolerans CF3咖啡因降解特性进行研究。通过检测降解率和生物量,确定了菌株CF3降解咖啡因最优条件为咖啡因质量浓度1 g/L、初始pH 4.5、培养温度30℃、转速190 r/min,在此条件下,咖啡因降解率可达99.5%。利用HPLC分离鉴定咖啡因降解物,在胞外培养液中检测到可可碱、副黄嘌呤和7-甲基黄嘌呤,因此确定菌株CF3是通过N-脱甲基途径降解咖啡因。
The caffeine degradation characteristics of Paraburkholderia caffeinitolerans CF3 strain screened and identified were studied by single factor test. The optimum conditions for the degradation of caffeine by strain CF3 were determined as follows: mass concentration of caffeine 1 g/L, initial pH 4.5, incubation temperature 30 ℃ and rotational speed 190 r/min. Under the condition, the degradation rate of caffeine could reach 99.5%. The degradation products of caffeine were separated and identified by HPLC. Theobromine, paraxanthine and 7-methylxanthine were detected in extracellular culture medium. Therefore, strain CF3 was determined to degrade caffeine through N-demethylation pathway.
