基于HPLC-Q-TOF/MS法分析马兜铃水煎剂化学成分的研究 被引量:5

摘要 目的运用高效液相串联四级杆飞行时间质谱(HPLC-Q-TOF/MS)法分析马兜铃水煎剂中的化学成分,为后续马兜铃的毒性研究提供借鉴。方法在总结马兜铃酸Ⅰ和马兜铃内酰胺Ⅰ质谱裂解规律的基础上,通过高分辨质谱提供的准确分子量及二级碎片离子信息,推测马兜铃药材中可能含有的化学成分,并对马兜铃酸类和马兜铃内酰胺类成分进行半定量分析。结果从马兜铃水煎剂中推测出18个化学成分,包括6个马兜铃酸类成分和7个马兜铃内酰胺类成分,其中马兜铃酸类成分的总峰面积是马兜铃内酰胺类成分总峰面积的1.23倍。结论尽管目前公认马兜铃酸Ⅰ和马兜铃酸Ⅱ是马兜铃的毒性化学基础,但是考虑到马兜铃中还含有很多其它马兜铃酸类成分以及马兜铃内酰胺类成分,这些成分也可能导致马兜铃药材的肝肾毒性,有待进一步深入的研究。这是首次用高分辨质谱分析鉴定马兜铃水煎剂中的马兜铃酸类和马兜铃内酰胺类成分,可为下一步马兜铃水煎剂成分的植化分离和毒性筛选提供借鉴。 Objective To provide references for the further toxicity study on Aristolochia contorta by identifying the chemical constituents in Aristolochia contorta water decoction, with utilizing high-performance liquid chromatography with quadrupole time-of-flight mass spectrometry(HPLC-Q-TOF/MS). Methods Based on summarizing the fragmentation patterns of aristolochic acid Ⅰ and aristolactam Ⅰ, the possible chemical ingredients in Aristolochia contorta water decoction with the help of accurate mass of molecular ion peaks and fragment ions provided by high-resolution mass spectrometry was detected. Meanwhile, we semi-quantified the content of aristolochic acid derivatives and aristolactam derivates in Aristolochia contorta water decoction. Results Consequently, there were 18 compounds tentatively identified in Aristolochia contorta water decoction,mainly including 6 aristolochic acid derivatives and 7 aristolactam derivates. The total peak areas of aristolochic acid derivatives were 1.23 times more than of those of aristolactam derivates. Conclusion Aristolochic acid Ⅰ and aristolochic acid Ⅱ are widely regarded as the chemical basis responsible for renal toxicity of Aristolochia contorta. However, there are many other aristolochic acid derivatives and aristolactam derivates, and those are also probably responsible for the hepatic and renal toxicity of Aristolochia contorta,awaiting further research. In short,this study is the first time to identify aristolochic acid derivatives and aristolactam derivates by high-resolution mass spectrometry in Aristolochia contorta water decoction, which may provide references for the further phytochemical separation and toxicity screening of Aristolochia contorta water decoction.
