摘要
目的比较不同培养基对骆驼刺内生细菌的分离效果,为内生细菌的分离培养选择合适的培养基。方法采集新疆荒漠地区骆驼刺植物种子,选用LB,TSA和R2A三种培养基对灭菌后的种子进行内生细菌分离纯化,并对内生细菌进行16S rDNA测序。结果从LB,TSA,R2A培养基上分别纯化到6、8、3株内生细菌,从TSA上分离到的内生细菌的数量和种类最多,与其他两种培养基比较有统计学意义(P<0.01)。芽孢杆菌属细菌是三种培养基上的优势菌群。结论TSA对于骆驼刺内生细菌的分离效果较好,是分离骆驼刺内生细菌较适宜的培养基。
Objective To compare the isolation effect of different culture media and choose the appropriate culture medium for endophytic bacteria of Alhagi.Methods Alhagi plant seeds were collected in Xinjiang desert region.Three kinds of culture media named LB,TSA and R2A were selected to isolate and purify endophytes after sterilization.Then 16S rDNA of endophytic bacteria were sequenced.Results Six,eight,and three strains of endophytic bacteria were purified from LB,TSA,and R2A media,respectively.The quantity and species of endophytic bacteria isolated from TSA were the most.The results were statistically significant compared with the other two media(P<0.01).Conclusion TSA has a better effect on the isolation of endophytic bacteria,so it is a suitable medium to isolate Alhagi endophytes.
