网络药理学探讨牛膝治疗股骨头坏死的可能分子机制 被引量:11

Discussion on the Possible Molecular Mechanism of Achyranthes Treatment for Femoral Head Necrosis based on Network Pharmacology

摘要 目的:运用网络药理学和生物信息学探讨牛膝治疗股骨头坏死的可能机制。方法:基于中药系统药理学数据库挖掘牛膝的活性成分,联合SwissTargetPrediction数据库对活性成分进行靶标预测,通过OMIM、TTD、GAD、GeneCards数据库检索与股骨头坏死相关的靶标,并与牛膝活性成分靶标映射明确牛膝治疗股骨头坏死的潜在靶标,运用Cytoscape3.6.0软件构建"药物-活性成分-潜在靶标"网络关系图。结合运用String数据库和Cytoscape3.6.0软件制作蛋白互作关系图。最后运用DAVID数据库对牛膝治疗股骨头坏死的靶标行通路富集分析。结果:从中药系统药理学数据库中共查询到牛膝成分176种,根据OB和DL参数筛选得到成分21种,删去无对应靶标的成分,最终得到活性成分17种。预测到牛膝活性成分的作用靶标456个。查询到与股骨头坏死发生相关的靶标82个。映射后得到牛膝治疗股骨头坏死潜在靶标38个。蛋白质互作关系图示:牛膝治疗股骨头坏死的关键靶标涉及VEGF、HIF1A、ABCB1等。DAVID富集显示:牛膝治疗股骨头坏死主要涉及Wnt/β-catenin、OPG/RANK/RANKL、HIF-1α、MAPK、TLR4/NF-KB等信号通路。结论:牛膝治疗股骨头坏死具有多成分、多靶点的作用特点,其主要通路参与调节骨代谢、调控血管生成、抑制炎症反应等控制疾病的发展。 Objective: Using network pharmacology and bio-informatics to explore the possible mechanism of Achyranthes treatment for femoral head necrosis. Methods: Based on the pharmacology database of traditional Chinese medicine system, the active components of Achyranthes bidentata were extracted, and the target was predicted by the Swiss Target Prediction database.Targets related to femoral head necrosis were searched by OMIM, TTD, GAD, Gene Cards database, and mapping with Achyranthes active target to identify potential targets for the treatment of femoral head necrosis by Achyranthes bidentata. Constructing a" drug-active ingredient-potential target" network diagram using Cytoscape 3.6.0 software. Combine the use of String database and Cytoscape 3.6.0 software to make protein interaction diagram. Finally, the DAVID database was used to analyze the pathological enrichment of the achyranthes in the treatment of femoral head necrosis.RESULTS: A total of 176 species of Achyranthes bidentata were obtained from the pharmacology database of Chinese medicine system. 21 components were selected according to OB and DL parameters,and the components without corresponding targets were deleted. Finally, 17 active components were obtained.456 targets were predicted for the active components of Achyranthes bidentata. 82 targets related to osteonecrosis of the femoral head were inquired. After mapping, 38 potential targets of Achyranthes bidentata for the treatment of femoral headnecrosis were obtained.The protein interaction diagram shows that the key targets of Achyranthes bidentata in the treatment of femoral head necrosis are vascular endothelial growth factor, HIF1 A, ABCB1, etc. DAVID enrichment showed that Achyranthes bidentata treatment of femoral head necrosis mainly involved in Wnt/beta-catenin, OPG/RANK/RANKL, HIF-1 a, MAPK, TLR4/NF-KB signaling pathways. Conclusion: Achyranthes bidentata has the characteristics of muti-component and muti-target in the treatment of femoral head necrosis. Its main pathway is involved in regulating bone metabolism, regulating angiogenesis and inhibiting inflammation to control the development of disease.
