川芎挥发油提取工艺优化及其地上部分GC-MS分析 被引量:14

Optimization of Extraction Process of Volatile Oil from Ligusticum chuanxiong and GC-MS Analysis of Its Overground Part

摘要 目的:采用Box-Behnken响应面优化川芎挥发油提取工艺,同时对川芎及其地上部分挥发油进行GC-MS对比分析。方法:RTX-5色谱柱(0.25 mm×30 m,0.25μm);载气为氦气;柱流速为1.0 mL/min;进样量1μL;进样口温度260℃;离子源温度200℃;柱温50℃,保持2 min,以5℃/min的速度升温至240℃并保持10 min;分流比40∶1;扫描方式为EI源;质谱标准库:NIST库。结果:川芎挥发油最佳提取工艺为:加10倍量水,浸泡时间2 h,提取时间9.19 h,挥发油提取率实测值为0.62%。经GC-MS分析,川芎饮片及其地上部分挥发油成分较为相似,但也存在差异,E-藁本内酯为其主要成分,含量分别占27.09%、23.11%。结论:该方法可用于川芎饮片及地上部分挥发油的成分分析,可为其地上部分开发及利用提供依据。 Objective:To optimize and analyse the volatile oil extraction process from Ligusticum chuanxiong and its overground part by using Box-Behnken response surface and GC-MS methods.Methods:With RTX-5 chromatographic column(0.25 mm×30 m,0.25μm),carrier gas was helium gas,flow rate was 1.0 mL/min,injection volume was 1μL,the split temperature was 260℃and ion source temperature was 200℃,column temperature was 50℃,keep 2 min and rising at a speed of 5℃/min to 240℃,keep 10 min,split ratio was 40∶1,scanning mode was EI source,MS standard library was NIST datebase.Results:The optimal extraction process of volatile oil from Ligusticum chuanxiong was as follows:the herb was ten times the amount of water,soaking time was 2 h,extraction time was 9.19 h.The measured value of extraction rate of volatile oil was 0.62%.Through GC-MS analysis,the compounds of volatile oil from Ligusticum chuanxiong decoction pieces and its overground part were similar,but there were some differences,E-ligustilide was its main component,the content accounted for 27.09%and 23.11%,respectively.Conclusion:The method can be used for the component analysis of volatile oil from Ligusticum chuanxiong decoction pieces and above ground.It can provide a basis for its development and utilization of above ground.
