对叶百部总生物碱止咳作用的谱效关系研究 被引量:17

摘要 目的:研究全国29个产地对叶百部总生物碱的HPLC图谱与止咳作用的相关性,建立各产地对叶百部总生物碱与止咳作用的谱效关系,阐明止咳作用的物质基础。方法:采用灰色关联度分析方法,对全国29个产地的对叶百部总生物碱的HPLC图谱的8个特征峰峰面积与止咳的2个评价指标(2 min内咳嗽次数、抑咳率)进行相关性分析,研究对叶百部总生物碱止咳作用的谱效关系。结果:对叶百部总生物碱对浓氨水致小鼠咳嗽有显著的抑制作用,与百部的止咳功效一致。在全国29个产地的对叶百部总生物碱HPLC图谱中,1号金刚大碱色谱峰峰面积大、关联度强;2~8号色谱峰对止咳作用也有较大的贡献,关联度差距较小,对止咳作用起协同作用。结论:全国29个产地的对叶百部总生物碱HPLC图谱与止咳作用具有相关性。通过谱效关系研究,可阐明对叶百部止咳作用的物质基础,为其质量控制与评价提供一定的理论依据。 Objective:To study the relationship between HPLC chromatogram and antitussive effect of total alkaloids from Stemona tuberosa and establish the spectrum-effect relationship between total alkaloids of Stemona tuberose from different regions and antitussive effect,to elucidate the material basis of cough-relieving effect.Methods:The grey relational analysis was used to analyze the correlation between the area of eight common peaks in the HPLC chromatogram and two evaluation indexes of cough relieving effect(coughing times in two minutes and cough inhibition rate)of total alkaloids from Stemona tuberosa samples collected in 29 producing areas in China,and the spectrum-effect relationship was studied.Results:The total alkaloids of Stemona tuberosa showed an obvious antitussive activity against concentrated ammonia water-induced cough in mice,which was consistent with cough suppression effect of Stemonae Radix.In the HPLC chromatogram of all twenty-nine samples,Croomine(peak 1)had a large peak area and high correlation with antitussive activity;peaks 2 to 8 also contributed a lot to the antitussive effect,with a small difference in correlation degree and a synergistic effect on the antitussive effect.Conclusion:The HPLC chromatogram and cough relieving effect of total alkaloids from Stemona tuberosa samples collected in twenty-nine areas in China are correlated.Through the study of spectral effect relationship,the material basis of antitussive effect of Stemona tuberosa is elucidated,which provids some theoretical basis for its quality control and evaluation.
