miR-8在脑膜瘤中的表达及抑制脑膜瘤细胞增殖的机制 被引量:1

摘要 目的探讨miR-8在脑膜瘤组织中的表达及对脑膜瘤细胞增殖能力的影响。方法采用原位杂交检测miR-8在脑膜瘤及瘤旁组织中的表达;脂质体转染使IOMM-Lee脑膜瘤细胞过表达miR-8,采用噻唑蓝(MTT)法、克隆形成实验检测IOMM-Lee增殖活性。检索TargetScan数据库预测miR-8的靶基因;双荧光素酶报告基因系统验证Ras相关的C3肉毒素底物(RAC)1是miR-8的靶基因。结果miR-8在脑膜瘤组织中的表达显著低于瘤旁组织(P<0.001);Ⅲ级脑膜瘤中miR-8的表达显著低于Ⅱ级、Ⅰ级(P<0.01);IOMM-Lee细胞过表达miR-8后,增殖抑制率、细胞克隆数目显著减少(P<0.05);TargetScan预测及双荧光素酶报告基因系统证实RAC1是miR-8的靶基因。结论miR-8通过与靶基因RAC1的3′UTR区结合,下调靶基因RAC1的表达,抑制脑膜瘤细胞增殖。
