摘要
目的 :研究自体静脉移植术后血浆内皮素 (ET)变化与移植静脉血管平滑肌细胞 (VSMC)增殖的关系。方法 :建立大鼠自体颈静脉移植于肾下腹主动脉动物模型 70只 ,于术后 3d～ 8周 ,采用放射免疫法检测血浆ET的变化 ,免疫组化法观察增殖细胞核抗原 (PCNA)在移植静脉VSMC中的表达和分布。结果 :术后 1～ 2周 ,血浆ET比术前明显增高 (P <0 .0 1) ;血浆ET与移植静脉VSMC增殖具有相关性 (r=0 .783,P <0 .0 1)。结论 :静脉移植术后测定血浆ET有助于反映移植静脉VSMC的增殖水平 ,为内膜增生的防治提供线索。
Objective: Our purpose was to study the change of plasma endothelin (ET) after vein grafting and their relationship with the proliferation of vascular smooth muscle cells (VSMC) in vein grafts. Methods: Autologous vein grafts were made by transplanting jugular vein to aorta in 70 Wistar rats. From 3 days to 8 weeks after grafting, plasma ET and proliferative cell nuclear antigen (PCNA) in VSMC of vein grafts were detected by using radioimmunoassay method and immunohistochemical method respectively. Results: Compared with normal control, the level of plasma ET from 1 to 2 weeks obviously increased (P<0.01). A significantly positive correlation existed between the level of ET and the proliferative rate (r=0.783, P<0.01). Conclusion: Level of plasma ET may indicate the proliferative activity of VSMC and give a clue for the treatment of intimal hyperplasia after vein grafting.
