铅对大鼠海马神经元胞质PKC活性的影响 被引量:5

摘要 目的 :观察不同低浓度醋酸铅不同染铅时间对大鼠海马神经元原代培养细胞胞质蛋白激酶C(PKC)活性及神经元细胞生长的影响。方法 :出生 4~ 8dWistar大鼠海马神经元原代细胞培养 ,显微照相 ,并不同时间不同浓度染铅 ,应用改良的Takai法测定PKC活性的变化。结果 :0 .1~ 1.0 μmol/L染铅 ,PKC活性依浓度依赖方式被激活 ,染铅浓度 >1.0 μmol/L ,PKC活性下降 ;染铅 15min后PKC活性升高并保持。低浓度醋酸铅并可抑制原代神经元细胞生长 ,呈浓度依赖方式。结论 :低浓度醋酸铅可持续激活大鼠海马神经元原代培养细胞胞质PKC ,抑制原代神经元细胞生长。 Objective: Our purpose was to observe how lead exposure of different low concentrations and time affects the activity of protein kinase C(PKC) in primary culture of hippocampal neuron of rats and neuron growth. Methods: The hippocampal neuron of Wistar rats which were 4 to 8 days old has been cultured. Experiment phase contrast microphotography and lead exposure of different concentrations and time on cell cultures have been used. The activity of cytosolic PKC was assayed by using improved Takai method. Results: Lead exposure of 0.1 to 1.0 μmol/L activated PKC in dose dependent manner. The activity of PKC decreased when lead concentration was more than 1.0 μmol/L. The activity of PKC reached the peak and persisted 15 minutes after lead exposure. Lead exposure of low concentration can inhibit neuron growth. Conclusion: Low level lead exposure can result in sustained activation of PKC and inhibit neuron growth in dose dependent manner.
