摘要
为探讨脂质过氧化能否诱导内皮细胞表达白细胞介素 - 1(IL - 1)及其是否具有对单核细胞的趋化活性 ,用不同浓度 (1、 5和 10 μmol/ L)的联胺诱发培养的内皮细胞脂质过氧化 ,用硫代巴比妥酸微量荧光测定法测定样品中的丙二醛含量 ;用免疫细胞化学检测 IL - 1β蛋白的表达 ;用单核细胞趋化实验观察脂质过氧化诱导的白细胞介素 - 1的趋化活性。结果显示 ,随着联胺浓度的增高 ,细胞内的丙二醛含量逐渐升高 ,分别为对照组的 2 .6 0倍、 5 .88倍和 11.6 7倍(F=12 9.10 ,P<0 .0 5 )。免疫细胞化学显示 ,正常内皮细胞可表达低水平 IL- 1β,加联胺后 ,各组 IL- 1β蛋白表达明显增加 ,分别为对照组的 1.78倍、 2 .70倍和 5 .0 2倍 (F=2 93.83,P<0 .0 5 )。趋化实验显示 ,经联胺刺激的条件培养基引起的单核细胞迁移距离明显大于非联胺条件培养基和随机移动组 (F=6 12 .5 5 ,P<0 .0 5 ) ,在联胺刺激的条件培养基中加入抗 IL - 1α或抗 IL - 1β抗体后 ,单核细胞迁移距离皆明显缩短 (F=4 37.0 3,P<0 .0 5 )。结果提示 ,联胺可诱发内皮细胞的脂质过氧化 ,诱导其表达和分泌 IL- 1,并初步证明 IL- 1可招引单核细胞迁移 ,从而在动脉内膜单核
The effect of lipid peroxidation on the expression of interleukin 1 (IL 1) in the cultured endothelial cells (ECs) was investigated and whether IL 1 is chemotactic for monocytes was explored. Lipid peroxidation of ECs was induced with 1 μmol/L, 5 μmol/L and 10 μmol/L diamide (DM) respectively, and the malondialdehyde (MDA) content in the ECs was determined by thiobarbituric acid flurophotometry. The expression of IL 1β protein was detected by immunocytochemistry, and the chemotactic activity of IL 1 was tested by monocyte chemotaxis assay. The results demonstrated that exposure of ECs to diamide at different concentrations mentioned above led to a 2 60 fold, 5 88 fold and 1 67 fold increase in MDA content as much as that of the control group respectively ( F=129 10, P <0 05). Immunochemistry showed that normal ECs could express IL 1β. Incubation of ECs to diamide at concentrations mentioned above resulted in a 1 78 , a 2 70 and a 5 02 fold increase in IL 1 content as much as that of the control group respectively ( F=293 83, P< 0 05). Monocyte chemotaxis assay revealed that the migration distance induced by diamide conditioned medium (DCM) was markedly longer than that of both non diamide conditioned medium (NDCM) group and random migration one (F=612 55, P<0 05) . After addition of anti IL 1β or anti IL 1 antibody into DCM, monocyte chemotactic activity was significantly decreased. (F=437 03, P<0 05). It was suggests that diamide can induce lipid peroxidation of ECs, which can promote ECs to express and secrete IL 1 and IL 1β protein, in turn, IL 1 might be chemotactic for peripheral blood monocytes and play an important role in the recruitment of monocytes into arterial intima during atherogenesis.
