摘要
用大孔网状树脂吸附法代替溶媒萃取法 ,分离纯化柱晶白霉素。从六种非极性树脂中确定国产CAD 45作吸附剂。确定了最佳提取工艺 :吸附最适 pH值为 8 5 ,流速 1/ 6 ( 1/min) ;解吸先用 1%氨水洗柱1∶1(v/v) ,流速 1/ 15 ( 1/min) ;采用丙酮解吸 ,收率可达 96 % ,成品效价 1346u/mg。
The macroreticular resin to adsorb leucomycin (LM) as the better method to replace the solvent extraction method was designed. The CAD-45 was screened as the adsorbent. The experiment result showed that the optimum pH value of adsorbent was 8.5, flow rate 1/6(1/min); the column was washed with 1% NH 4OH, 1∶1(v/v), at a flow rate of 1/15(1/min). The yield can increased to 96% with Me 2CO as the eluent and the potency of the product was 1346u/mg.
