摘要
以吸水链霉菌应城变种10-22菌株为指示菌,从土壤中分离得两个温和性噬菌体,φHAU3和φHAU4。多次单斑分离纯化证明φHAU3的噬菌斑为小斑,φHAU4为大斑;SalGⅠ酶切分析结果表明此两株噬菌体DNA酶切片段完全不同,分属两个不同的噬菌体;它们的寄主范围很广,在49个菌株(分属于7个属,38个种)中,φHAU3可以感染28个,φHAU4可以感染34个;均可感染链霉菌属以外另外两个属的放线菌菌株——地中海拟无枝酸菌和吸水孢囊放线菌;不能感染限制性内切酶SalGⅡ产生菌——白色链霉菌G,但可以感染其限制酶缺陷型突变株J1074。两株噬菌体的pH适应范围都较广(在pH4~9之间);pH4时φHAU3存活率为96.6%,φHAU4为79.9%;pH达到10时,两株噬菌体的存活率都降至40%;pH11时全部致死。最适温度均为28~37℃,50℃处理30分钟φHAU3成活率仍为57%,而φHAU4则只有9%。用37种限制性内切酶对两种噬菌体DNA进行了单酶切分析,发现它们的DNA均为双链,也均具有粘性末端。两个噬菌体DNA的限制性内切酶片段之和表明,φHAU3和φHAU4的分子量均在50kb左右。两个噬菌体的DNA可转染变铅青链霉菌,但尚不能转染10-22菌株。
Two temperate phages,φHAU3 and φHAU4,were isolated from soil using Streptomyces hygroscopicus 10-22 as indicator strain. The plaques formed by φHAU3 seemed to be much smaller than those formed by φHAU4. These two phages are apparently different because they showed very different banding patterns on agarose gel after digestion with SalG I. Both phages showed very wide host-range. While φHAU3 can infect 29 out of 49 different actinomycete strains (belong to 7 genera and 38 species), φHAU4 can infect 34 of the 49 strains. None of the two phages can infect S.al bus G ( SalG I producer ) but both of them can infect its restriction minus mutant J1074. Both phages can infect two representative strains of Amycolatopsis mediterranei and Actinosporangium hygroscopicus, which are outside of Streptomyces genus. Plaques formed by φHAU4 seemed to be bigger than those formed by φHAU3. Both phages are stable at the pH range between 4-9, but φHAU3 is more acid resistant. Furthermore, φHAU3 is more heat-resistant than φHAU4. After treatment at 50℃ for 30 min, 57% of φHAU3 remains viable, but only 9% of φHAU4 are still alive. The suitable incubation temperature for both phages are between 28-37℃. DNAs of φHAU3 and φHAU4 were digested by 37 restriction enzymes giving rise to different number of restriction fragments, indicating that their DNAs are both double-stranded. Both phage genomes contain cohesive ends judging from their slightly different banding patterns of the Bgl I -digested DNAs before and after their treatment at 70℃. The sum of the restriction fragments of each phage genome are about 50 kb. The DNAs isolated from both phage can transfect S.I ivdans, but not S. hygroscopicus 10-22.
