摘要
从鸭乙型肝炎病毒(DHBV)DNA阳性鸭的外周血分离单个核(PBMN)细胞,洗涤10次后,腹腔接种出壳24小时以内的雏鸭。接种量为每只2.5×10~6个细胞。接种后每隔2周取血检测血清中鸭乙型肝炎病毒表面抗原(DHBsAg)及DHBV DNA。在分别两次试验中发现,接种后2周9%~50%受体鸭转为DHBV DNA阳性,至第6周阳性者中大部分鸭DHBV DNA仍持续阳性。以末次细胞洗涤液及DHBV DNA阴性鸭的PBMN细胞分别接种雏鸭作对照,均未致感染。本文首次证明DHBV感染鸭的用聚蔗糖-泛影葡胺分离提取的单个核细胞具有感染性。
Peripheral blood mononuclear cells ( PBMN ) from DHBV persistently infected ducks ( DHBV positive ) were isolated, washed for 10 times and inoculated intraperitoneally into ducklings 24 hours after hatching. A total number of 2.5×108 cells were inoculated and sera from recipient ducklings were assayed for DHBV DNA as well as for DHBsAg every 2 weeks up to 6 weeks. It was found that after 2 weeks,9%-50% of the recipient ducklings were seroconverted into DHBV DNA and DHBsAg positive in two separate studies. Up to 6 weeks after inoculation, some ducks were still positive, PBMN from a DHBV DNA negative duck ( normal duck) and the supernatent of the last wash fluid of PBMN from DHBV DNA positive ducks, when inoculated into duckling, did not cause infection. Data of this report indicate that PBMN from infected ducks are infectious, though the mechanisms of infection still merits further study.
