摘要
用荧光及CD光谱研究了顺铂与人红细胞收缩蛋白(SP)的作用。顺铂可使SP的内源性荧光相对强度改变。SP的巯基和氨基是顺铂的结合基因,顺铂与SP的反应遵从双阶段一级反应动力学,24℃下k_1=9.28×10^(-3)分^(-1),k_2=2.86×10^(-3)分^(-1)。顺铂可引起SP的构象变化,这种变化与顺铂浓度有关。
The interaction between cisplatin and spectrin from human erythrocytes were studied by fluoresce-nce spectroscopy. Cisplatin can change the relative strength of the intrinsic fluorescence of spectrin.Both the mercapto and amino grouos from spectrin were shown binding to the platinum by means offluorescence labeling technique. The reaction of cisplatin and spectrin follows a two-step first orderreaction. The rate constants K_1 and K_2 were determined to be 9.28×10^(-3) min^(-1) and 2.85×10^(-3) min^(-1)respectively. It has been confirmed that cisplatin can cause conformation change of spectrin with CDspectra. This change is dependent on the concentration of cisplatin.
