大鼠脑缺血后胶质细胞向树突状细胞转化的研究 被引量:6

Study on differentiation of microglia cells to dendritic cells after middle cerebral artery ischemia in rat

摘要 目的 探讨缺血脑组织中树突状细胞 (DC)的来源 ,以其及是否参与脑缺血损伤的过程。方法 用线栓方法封闭大鼠右侧大脑中动脉制作动物模型。用免疫组化染色法 ,检测脑缺血 1h到第 6天时 ,缺血脑组织中DC(OX6 2 + )的存在 ,以及胶质细胞 /巨噬细胞 (OX4 2 + )转化成DC(OX6 2 + )的情况。同时检测活化后的DC样细胞表达MHC Ⅱ类分子的水平。结果 缺血脑半球和假手术的脑半球相比较 ,在 12hDC的数量明显增加 (P <0 .0 0 1)。在脑缺血组中 ,缺血脑半球与非缺血脑半球相比较 ,DC的数量也增多 (P <0 .0 0 1)。脑缺血第 6天 ,缺血组与假手术组进行比较 ,DC表达MHC Ⅱ类分子 (OX6 2 + OX6 + )显著升高(P <0 .0 0 1)。缺血脑半球在 1h到第 6天 ,OX4 2 + 的细胞转变成OX6 2 + OX4 2 + 的细胞逐渐增加 ,第 6天缺血脑半球与非缺血脑半球相比较显著增多 (P <0 .0 0 1)。脑缺血损伤的面积与以每 10 0mm2 脑组织切片为单位的OX6 2 + 细胞的数量呈正相关 (R2 =0 .8914 ,P <0 .0 0 1)。结论 脑缺血后 ,脑缺血组织中DC数量的增加与脑损伤的面积呈正相关 ,提示DC参与了脑缺血过程。大鼠脑缺血后 ,从胶质细胞向DC样细胞的转化是缺血脑组织中DC的来源之一。 Aim To explore the source of dendritic cells (DC) in schemic brain tissue and its whether participate in the injury process of cerebral ischemia. Methods The right middle cerebral arteryies (MCA) of all experimental rats were occluded by inserting a nylon monofilament in the right external carotid artery. Immunohistochemical staining was used to detect DC (OX62 +), microglia/macrophage (OX42 +) that transformed into DCs, and expression of MHC Ⅱ molecule on activated DC like cells in ischemic brain hemispheres at 1 hour to 6th day after middle cerebral ischemic occlusion (MCAO) or shamming operation. Results The number of DCs in ischemic brain hemisphere was notably more than that in shamming operation group ( P <0.001) at 12 hours after MCAO. Expression of MHC Ⅱmolecule (OX62 + and OX6 +) on DCs in ischemic brain tissue at 6th day after MCAO also markedly increased as compared with shamming operation group ( P <0.001), and with prolongation of ischemic time, the number of DCs expressing OX62 and OX6 which transformed by OX42 + cells increased gradually. The area of ischemic brain lesion correlated directy to the number of OX62 + cells in per 100 mm 2 of brain tissue section ( R 2=0.8914, P <0.001). Conclusion After ischemic brain lesion, the increased number of DCs suggests that DCs have participate in brain ischemia pocess, and DC like cells mainly stem from microglia in the ischemic brain tissue.
