摘要
以体外培养瘤细胞集落形成率、荷瘤小鼠生命延长率(ILS)和小鼠实体瘤的抑瘤率作为药效的评价指标,观察顺铂(DDP)与阿霉素(ADM)合用的剂量效应关系。小剂量DDP与ADM的联用时相加作用明显。一种药已达有效量再与另一药小剂量合用其效应与单药足量的效应无显著差异;若两药有效量联合,虽能提高ILS,但表现出毒性增强。
By the determination of colony formation rate of tumor cells in culture,the increase percentage in life -span (%) of tumor-bearing mice, and solid tumor growth in-hibitizon rate in mice, the dose-response relationship of cisplatin (DDP) and adriamycin (AD-M) combination therapy was evaluated in experiments in vivo and in vitro. DDP and ADM combination therapy showed additive effect on the antitumor activity. The obvious additive effect occured in bwer dose of these two drugs in combination. If the dose of one drug 1/4 of the effective dose combined with ineffective dose (1/16, 1/64 of effective dose) of another drug, the colony formation rate was decreassd obviously, but when the effective dose (DDP and 80 n mol/ml for DDP or ADM) of one drug was combined with a sub-effective dose of another drug,the colony formation rate did not deoease.The dose relationship in vivo was similar to that in vitro. The combination of ineffective doses (0.08, 0.25 or 0.05 mg/kg X 4 ) of two drugs may increase the value of ILS of mice bearing EAC to 215.6% (P<0.01), it was dose-dependent When one drug was used in effective dose combined with the ineffective dose of another drug, the value of ILS did not increase obviously (P>0.05), but the toxicity was increased. These results suggested that in dinical combination drug therapy the dose of single drug must be reduced,otherwise would be harmful
