猪肠道中抑制大肠杆菌的乳酸菌的分离与筛选 被引量:12

摘要 采用平板稀释法 ,用 MRS和 M17培养基从 12头猪的新鲜粪便中分离出 2 0 0个菌株 ,进而初筛得 38株对普通大肠杆菌有抑制作用、复筛得 10株对猪源弱毒型大肠杆菌有较强抑制作用、再选出其中抑菌活性最强的 3株 R- 2 1- 1、R- 17- 3和 R- 2 1- 3。生理生化特性试验表明这 3株均为乳酸菌。以多粘菌素 B对弱毒型大肠杆菌抑制作用所得的标准曲线作为参照 ,测得这 3株菌的抑菌活性分别相当于 12 80 0 ,135 0 0和 30 2 0 IU.m L-1的多粘菌素 B,蛋白酶试验、排除酸等试验判断其产生的抑菌物质中含有细菌素 ;通过抑菌谱试验证明 3株菌不仅对革兰氏阳性的单核细胞增生利斯特氏菌和金黄色葡萄球菌有明显的抑制作用 ,而且对革兰氏阴性菌的猪霍乱沙门氏菌和鸡白痢沙门氏菌有明显的抑制作用 ; Two hundred strains of microorganism were isolated from fresh feces of 12 pigs of different ages. Thirty eight strains can inhibit the growth of Escherichia coli. Ten strains can inhibit the growth of enterotoxingenic E.coli (C83917). Three of them are most effective (Test proved that they are all lactic acid bacteria and were named R 21 1, R 21 3 and R 17 3.). A standard curve was established by using polymyxin B which can also inhibit the growth of enterotoxingenic Escherichia coli . Refering to this curve, the bacteriostatic activity of R 21 1, R 17 3 and R 21 3 are 12800, 10350, 3020IU·mL -1 respectively. Their bacteriostatic products were improved to contain bacteriocin. The test of bacterial inhibition zone proved that the three LAB can inhibit not only positive bacteria (Gram stain) such as staphylococcus aureus and Listera monocytogenes, but also negative bacteria as E.coli, Salomonella (C500) and Salmonella (C97 13).
