摘要
来乐司酮是抗孕激素催经止孕药RU_(486)的类似物,其特点是动物实验的抗早孕作用比RU_(486)强,但抗糖皮质激毒活性却比后者低,是一个有希望成为RU_(486)的换代药。Ames、染色体畸变及小鼠骨髓多染红细胞微核计数等三项致突变试验结果表明,来乐司酮均为阴性。
Lilopristone(ZK98734)is a derivative of RU486.In order to check its safety, a group of mutagenic tests including Ames test, chromosome aberration assay in CHO cell culture and micronucleous test in mice has been carried out.Home-made ZK98734 proeuced no mutagenic effect both in the Ames test up to 5mg per dish on 4 strains(TA97, TA88, TA100, and TA102)and in the chromosome aberration assay with 40ug/ml on the CHO cell, with or without metabolic activation.Home-made ZK98734 did not change the incidence of micronucleated polychromatic erythrocytes in the bone marrow of mice, either 24, 48, or'72 hours after oral treatment with 1.5g/kg.
