摘要
目的以透明质酸(hyaluronic acid,HA)对介孔二氧化硅纳米颗粒进行功能化(mesoporous silicana noparticle,MSN),并负载抗肿瘤药物顺铂(cisplatin,CDDP),制备CDDP@HA-MSN纳米药物,研究其形貌特征以及对人乳腺癌细胞(MDA-MB-231)的杀伤作用。方法通过扫描电镜(TEM)和动态光散射粒径分布仪(DLS)表征其形貌,通过MTT法检测其对MDA-MB-231细胞的杀伤作用。结果CDDP@HA-MSN平均粒径为150nm,载药量为18.7%,具有透明质酸酶响应性,细胞实验结果表明该纳米药物可明显增强对MDA-MB-231细胞的杀伤效果。结论成功制备了纳米药物CDDP@HA-MSN,能够显著增强抗肿瘤作用,有望提高对乳腺癌的靶向治疗效果。
Objective:To prepare the CDDP@HA-MSN nanomedicine by functionalizing Mesoporous Silica Nanoparticles(MSN)with hyaluronic acid(HA)and being loaded with cisplatin(CDDP),and to study the morphology and the effects on the survival rate of human breast cancer cells(MDA-MB-231).Methods:Its morphology was characterized by TEM and DLS,the proliferation inhibition of MDA-MB-231 cells was detected by MTT assay.Results:The average size of CDDP@HA-MSN nanoparticles was about 140 nm,and the drug load was 18.7%.The results of cell experiments showed that the nanomedicine significantly inhibited the proliferation of MDA-MB-231 cells.Conclusion:CDDP@HA-MSN can significantly enhance the killing ability of tumor cells,and it is expected to improve the efficacy of chemotherapy for breast cancer.
