摘要
从江西九连山霉烂毛红椿种子上分离种腐病原菌,采用形态学和分子生物学方法对其进行鉴定,并采用菌落直径法研究其在不同培养基、温度、pH以及多种碳氮源条件下的生物学特性。结果表明:从霉烂毛红椿种子上分离到一株对毛红椿种子具有强致病性(编号为JZF02)的真菌,经鉴定为厚垣镰刀菌(Fusarium chlamydosporum);该菌菌丝在不同培养基上的生长速率表现为PSA>PDA/MA>CA>CD;该菌能利用多种碳氮源,但铵盐不适合该菌生长;在0~40℃均能生长,最适生长温度为25~30℃;在pH5~12条件下均能生长,其中最适pH为8。本研究为掌握厚垣镰刀菌引起的毛红椿种子病害发生规律及采取防控措施提供了理论依据。
In this study,the pathogen isolated from the rotten seeds of Toona ciliata var. pubescens collected from Jiulianshan Mountains of Jiangxi Province and the morphological and molecular methods were used to identified the pathogen.And the biological characteristics of T. ciliata pathogen in different medium,temperature,pH and various carbon and nitrogen sources were studied by colony diameter method.The results showed that a fungus with strong pathogenicity (no.JZF 02) was isolated from mouldy T. ciliata seeds and identified as Fusarium chlamydosporum;The growth rate of fungal hyphae on different media showed PSA > PDA/MA > CA > CD;The strain could use a variety of carbon and nitrogen sources,but ammonium salt was not suitable for the growth of the bacteria;It could also grow at 0-40 ℃,and the optimum growth temperature was 25-30 ℃;Additionally,it could grow at pH 5-12,and the optimum pH was 8.This study provided a theoretical basis for mastering the occurrence rules of seed diseases caused by F. tabacum and taking prevention and control measures.
