摘要
目的探讨促红细胞生成素对新生大鼠高胆红素血症的脑保护作用。方法7 d龄清洁级SD雄性新生大鼠60只分成3组,模型组、对照组与EPO组,每组各20只。所有大鼠均建立高胆红素血症模型,对照组在模型建立后即刻给予腹腔注射鼠神经生长因子,EPO组在对照组基础上经颈静脉注射EPO,模型组注射同剂量的PBS缓冲液,1次/d,共3 d。结果EPO组与对照组治疗后1 d、2 d与3 d的Tarlov评分高于模型组(P<0.05),EPO组也高于对照组(P<0.05)。治疗后3 d,EPO组海马组织的SOD含量高于对照组与模型组(P<0.05),MDA含量低于对照组与模型组(P<0.05),对照组与模型组对比差异也有统计学意义(P<0.05);EPO组的IL-6和TNF-α值低于对照组与模型组(P<0.05),对照组低于模型组(P<0.05)。结论EPO在新生大鼠高胆红素血症中的应用能抑制炎症因子的表达,改善氧化应激反应,促进大鼠神经行为功能的恢复,从而发挥脑保护作用。
Objective To investigate the protective effect of erythropoietin(EPO)on hyperbilirubinemia in neonatal rats.Methods 60 male SD rats of 7-day-old clean grade were divided into three groups:model group,control group and EPO group,with 20 rats in each group.All rats were established with hyperbilirubinemia model.The control group were given intraperitoneal injection of mouse nerve growth factor immediately after the model were established,the EPO group were injected with EPO via the ear vein on the basis of the control group,and the model group were injected with the same dose of PBS,1 time/d for 3d.Results The Tarlov scores of the EPO group and the control group were higher than those of the model group in the 1 day after treatment,2 days after treatment and 3 days after treatment(P<0.05),and the EPO group were also higher than the control group(P<0.05).At 3 days after treatment,the SOD content in hippocampus of EPO group were higher than that of control group and model group(P<0.05),MDA content were lower than that of control group and model group(P<0.05),and the difference compared between control group and model group were also statistically significant(P<0.05).The IL-6 and TNF-a values in the EPO group were lower than those in the control group and the model group(P<0.05),and the control group were lower than the model group(P<0.05).Conclusion The application of EPO in hyperbilirubinemia in neonatal rats can inhibit the expression of inflammatory factors,improve oxidative stress,promote the recovery of neurobehavioral function in rats,and thus plays a protective role in the brain.
