摘要
目的基于网络药理学研究泽泻汤治疗脑水肿的作用机制。方法检索泽泻汤中药物成分,预测相应靶点,将靶点转换为相应基因。Cytoscape3.2.1中构建泽泻汤有效成分-靶点网络;检索脑水肿相关基因,Cytoscape3.2.1中构建脑水肿疾病靶点网络;Cytoscape3.2.1中将泽泻汤有效活性成分-靶点网络和疾病靶点网络合并;进行GO功能和k EGG通路富集分析。结果 GO功能分析结果为平滑肌细胞迁移、细胞周期过渡正向调节、DNA损伤检查点、DNA损伤反应、p53类介质的信号转导造成细胞周期停滞、转录起始调控、应激反应中起始转录的调控、转录因子进入细胞核调控、核内蛋白质的正向调节、核质转运的正向调节,k EGG富集通路主要为Erb B信号通路、v EGF信号通路。结论泽泻汤的有效活性成分可能通过上调p53基因表达率、阻断v EGF通路、阻断Erb B信号通路、阻断Erb B/v EGF信号通路缓解脑水肿。
Objective To explore the mechanism of Zexie Decoction in treating glioma cerebral edema based on network pharmacology. Methods Firstly, retrieving the herbal ingredients of Zexie Decoction and predicting the corresponding target point, and then convert the corresponding gene. And then, importing effective active ingredients and target corresponding genes into Cytoscape3.2.1 software to build an effective active component of Zexie Decoction-target network. Secondly, retrieving the genes related to glioma cerebral edema, and import them into Cytoscape3.2.1, then constructing the disease-target PPI network of glioma cerebral edema. Thirdly, merging the effective active component of Alisma Soup-target PPI Network and the disease-target network of glioma cerebral edema by the Cytoscape3.2.1. Lastly, analyzing the GO function and the enrichment of kEGG pathway. Results The results of GO function analysis are smooth muscle cell migration, positive regulation of cell cycle phase transition, DNA damage checkpoint, DNA damage response, signal transduction by p53 class mediator resulting in cell cycle arrest, regulation of translational initiation, regulation of translational initiation in response to stress, regulation of transcription factor import into nucleus, positive regulation of protein localization to nucleus, positive regulation of nucleocytoplasmic transport. The enrichment pathway of kEGG is mainly the function of p53 gene, ErbB signaling pathway and vEGF signaling pathway. Conclusion The effective active component of Alisma soup may relieve glioma brain edema by increasing the p53 gene expression rate, blocking the vEGF pathway, blocking the ErbB signaling pathway, and blocking the ErbB/vEGF signaling pathway.
