摘要 目的观察ADAM12L在胃癌中的表达及临床意义。方法96例胃癌患者,qRT-PCR测定ADAM12LmRNA表达情况,Westernblot方法检测蛋白的表达,分析其与胃癌临床病理特征及预后之间的关系,多因素Cox回归分析预测不良事件的指标。结果胃癌组织的ADAM12LmRNA表达水平明显高于癌旁组织(P<0.05),癌组织的ADAM12L蛋白表达水平明显高于癌旁组织(P<0.05),与组织学分级、临床分期、淋巴结转移、浸润程度存在一定的相关性(P<0.05)。ADAM12L基因高表达组和低表达组整体生存曲线分布比较,差异有统计学意义(P<0.05)。组织分学分级、临床分期、淋巴结转移、浸润程度和ADAM12L水平是重要的预测不良事件的指标。结论ADAM12L无论是在mRNA水平还是蛋白质水平,其在胃癌组织表达均上调,而且其上调与胃癌的发生、进展以及临床预后可能密切相关。 Objective To observe the expression and clinical significance of ADAM12L gene in gastric cancer.Methods The expression of ADAM12L mRNA was detected by qRT-PCR in 96 patients with gastric cancer.Its expression of protein was detected by Western blot,and its relationship was analyzed between clinicopathological features and prognosis of gastric cancer.Multivariate Cox regression analysis was used to predict indicators of adverse events.Results The expression of ADAM12L mRNA in cancer tissues was significantly higher than that in para-carcinoma tissues(P<0.05),the expression level of ADAM12L protein in cancer tissues was significantly higher than that in para-carcinoma tissues(P<0.05),which showed certain correlation with histological grade,clinical stage,lymph node metastasis,and degree of infiltration(P<0.05).The distribution of overall survival curves between high and low expression group were statistically significant(P<0.05).Tissue grade,clinical stage,lymph node metastasis,degree of infiltration,and ADAM12L levels were important indicators of predictive adverse events.Conclusion Both mRNA level and the protein level of ADAM12L were up-regulated in gastric cancer tissues,which may be closely related to the occurrence,progression and clinical prognosis of gastric cancer.
