摘要
目的研究MitoQ对缺氧PC12细胞的保护作用。方法建立PC12细胞缺氧模型,通过观察细胞形态,检测细胞活性、氧化应激、脂质过氧化、线粒体功能相关指标以及凋亡相关蛋白表达,探讨MitoQ对缺氧PC12细胞的保护作用。结果与模型组相比较,MitoQ能显著提高缺氧PC12细胞活性(P<0.01),降低缺氧PC12细胞活性氧、丙二醛、钙离子浓度(P<0.05)、提高线粒体膜电位(P<0.01),显著降低缺氧条件下PC12细胞凋亡蛋白Bax的表达(P<0.01)。结论缺氧引起PC12细胞凋亡,MitoQ通过降低细胞氧化应激、脂质过氧化水平,提高缺氧细胞线粒体功能,对缺氧PC12发挥保护作用。
Objective To study effect of MitoQ in protection of hypoxia injury in PC12 cells.MethodsAnoxic models of PC12 cells were established.Cell appearances were observed,and cell activity,oxidative stress,lipid peroxidation,related indicators of mitochondrial function and expressions of apoptosis related proteins were detected to study protective effects of MitoQ on hypoxia injury in PC12 cells.ResultsCompared with those in control group,MitoQ could significantly improve PC12 activity(P<0.01),and it also could significantly reduce concentrations of reactive oxygen species(ROS),malondialdehyde(MDA)and Ga 2+of hypoxia PC12 cells(P<0.05),increase mitochondrial membrane potentials(P<0.01)and significantly decrease expression of PC12 apoptosis protein Bax under hypoxia condition(P<0.01).ConclusionAnoxia may induce PC12 apoptosis,and MitoQ may protect hypoxia injury in PC12 cells by reducing levels of oxidative stress and lipid peroxidation and increasing mitochondrial function.
