CDKN2A/2B基因单核苷酸多态性与妊娠期糖尿病的相关性 被引量:2

Correlation of CDKN2A/2B gene single nucleotide polymorphism with gestational diabetes mellitus

摘要 目的探讨细胞周期素依赖性蛋白激酶抑制因子2A/2B(CDKN2A/2B)基因单核苷酸多态性(SNP)与妊娠期糖尿病(GDM)发生风险的关系。方法选择459例GDM患者为病例组,571例正常妊娠者为对照组。通过相关数据库及文献筛选CDKN2A/2B基因的7个候选SNP位点,并对其进行基因分型。采用多因素Logistic回归模型评估候选SNP位点与GDM的相关性,采用多因子降维法评估候选SNP位点的单独或联合作用对妊娠期女性发生GDM的影响。结果多因素回归分析结果显示,与携带CDKN2A/2B基因rs10811661位点CC基因型的孕妇比较,携带TC或TT基因型的孕妇GDM患病风险升高(均P<0.05);在显性遗传模型下,与CC基因型携带者比较,携带TC/TT基因型的孕妇发生GDM的风险升高(P<0.05)。多因子降维分析结果显示,最优单位点模型为rs10811661位点(P<0.05);SNP-SNP联合作用与GDM发病存在关联,rs10811661-rs7036656-rs1063192-rs3217986位点的交互作用可增加GDM发病风险(P<0.05)。结论 CDKN2A/2B基因rs10811661位点多态性与GDM发生有关,SNP单独和联合作用均可影响妊娠期女性对GDM的易感性。 Objective To explore the association between the single nucleotide polymorphism(SNP) of cyclin -dependent kinase inhibitor 2A/2B(CDKN2A/2B) gene and the risk of developing gestational diabetes mellitus(GDM). Methods A total of 459 patients with GDM were enrolled as case group,and 571 women with normal pregnancy as control group.Seven candidate SNP loci of CDKN2A/2B gene were screened out from related databases and literatures,in which genotyping was performed.Multivariate Logistic regression model was employed to assess the correlation of candidate SNPs with the risk of developing GDM,multifactor dimensionality reduction method was used to assess the single or combined effects of candidate SNP loci on the incidence of GDM in pregnant women. Results Multivariate regression analysis results showed that,compared with pregnant women with genotype CC at locus rs10811661 of CDKN2A/2B gene,those with genotype TC or TT were at a higher risk of suffering GDM(all P <0.05);in the model of dominant inheritance,pregnant women with genotype TC/TT were at a higher risk of developing GDM compared with those with genotype CC( P <0.05). Multifactor dimensionality reduction analysis results showed that locus rs10811661 was the optimal single-locus model( P <0.05);the combined effect of SNP-SNP was linked to the incidence of GDM, interaction of loci rs10811661-rs7036656-rs1063192-rs3217986 could increase the risk of developing GDM( P <0.05). Conclusion CDKN2A/2B gene locus rs10811661 polymorphism is associated with the incidence of GDM,and single or combined effects of SNP might affect the susceptibility of pregnant women to GDM.
