摘要
目的制备姜黄素固体分散体单层渗透泵控释片,并进行处方优化。方法溶剂法制备固体分散体后,再进一步制成单层渗透泵控释片。以NaCl用量、CMC-Na用量、PEG 4000用量、包衣增重为影响因素,累积释放度为评价指标,正交试验优化处方。然后,考察其体外释药行为。结果最佳处方为NaCl用量170 mg,CMC-Na用量25 mg,PEG 4000用量12%,包衣增重4.5%。单层渗透泵控释片释药速率恒定,12 h内累积释放度达92.42%,符合零级模型(r=0.992 6)。结论该方法稳定可靠,可用于制备姜黄素固体分散体单层渗透泵控释片。
AIM To prepare the single-layer osmotic pump controlled release tablets of curcumin solid dispersions and to make formulation optimization.METHODS The solid dispersions were prepared by solvent method, and then further prepared into single-layer osmotic pump controlled release tablets. With NaCl consumption, CMC-Na consumption, PEG 4000 consumption and coating weight gain as influencing factors, accumulated release rate as an evaluation index, the formulation was optimized by orthogonal test. Subsequently, the in vitro drug release behaviors were investigated.RESULTS The optimal formulation was determined to be 170 mg for NaCl consumption, 25 mg for CMC-Na consumption, 12% for PEG 4000 consumption, and 4.5% for coating weight gain. The single-layer osmotic pump controlled release tablets demonstrated constant drug release rate with the accumulated release rate of 92.42% within 12 h, which accorded with zero-order model.CONCLUSION This stable and reliable method can be used for preparing the single-layer osmotic pump controlled release tablets of curcumin solid dispersions.
