摘要
为从红树林微生物中分离抗肿瘤活性物质,从泉州湾红树土壤中分离、纯化和筛选能产生抗肿瘤活性物质的细菌,并利用16S r DNA序列比对方法进行分子鉴定.采用追踪分离的方式,以稻瘟霉分生孢子抑制强度和肿瘤细胞体外抑制活性为指标,对细菌发酵液抗肿瘤活性成分进行初步分离.实验结果显示,从泉州湾红树林土壤中分离出1株具有较强抗肿瘤活性的细菌,经鉴定为Vibrio fluvialis. Vibrio fluvialis发酵液能抑制稻瘟霉分生孢子萌发时菌丝形变,并抑制EC9706食管癌细胞和H22肝癌细胞生长.从Vibrio fluvialis发酵液分离出具有较强抗肿瘤活性的化学组分Ⅰ和组分Ⅱ.组分Ⅰ对EC9706食管癌细胞和H22肝癌细胞ρIC50分别为0. 18 mg·m L^-1和0. 26 mg·m L^-1,组分Ⅱ对EC9706食管癌细胞和H22肝癌细胞ρIC50分别为0. 17 mg·m L^-1和0. 22 mg·m L^-1.表明Vibrio fluvialis能产生抗肿瘤活性物质,具有重要抗肿瘤研发价值.
To isolate anti-tumor active substances from mangrove microorganisms,an antitumor bacterium was isolated and identified using phylogenetic analysis of 16S rRNA gene sequences. Extraction and preliminarily isolation of the anti-tumor substances from fermented broth of this bacterium were studied using bioassay-guided fractionation. The anti-tumor bacterium successfully isolated from mangrove soil of Quanzhou bay was identified as Vibrio fluvialis. The fermented broth of Vibrio fluvialis could induce deformation of mycelia of Pyricularia oryzae and inhibit the growth of EC9706 esophageal cancer cells and H22 liver cancer cells. Two anti-tumor chemical components(Ⅰand Ⅱ) were isolated from fermented broth of Vibrio fluvialis. The 50% concentration of inhibition(ρIC50) of EC9706 esophageal cancer cells and H22 liver cancer cells were 0. 18 mg·m L^-1 and 0. 26 mg·m L^-1,respectively. The ρIC50 of EC9706 esophageal cancer cells and H22 liver cancer cells were 0. 17 mg·m L^-1 and 0. 22 mg·m L^-1,respectively. The active component separated from the fermented broth of Vibrio fluvialis has a significant antitumor activity,which was worth exploiting as potential antitumor drugs.
