基于Oncomine数据库分析ARID5B基因在卵巢癌中的表达意义及血根碱的干预研究 被引量:5

摘要 目的:通过挖掘Oncomine数据库中的相关数据,分析ARID5B基因在卵巢癌中的表达及临床意义,并探讨血根碱对卵巢癌细胞中ARID5B基因的干预作用。方法:收集Oncomine数据库中关于ARID5B研究的信息,对其在卵巢癌的水平变化进行荟萃分析,采用Kaplan-Meier法分析ARID5B水平与卵巢癌患者生存时间之间的关系,探讨其临床意义。卵巢癌SKOV3细胞,分为对照组和血根碱组,48小时后提取总RNA,用全基因芯片检测基因表达谱,筛选差异基因,探讨血根碱对卵巢癌SKOV3细胞ARID5B基因表达的影响。结果:分析Oncomine数据库中收集的卵巢癌和正常卵巢组织ARID5B基因水平的13项研究数据,发现卵巢癌组织中ARID5B基因的表达显著低于正常卵巢组织(P<0.05)。通过生存分析发现低表达ARID5B基因的卵巢癌患者无病生存期(PFS)、总生存期(OS)明显长于高表达者,高表达患者的预后更差(P<0.05)。血根碱显著上调卵巢癌SKOV3细胞ARID5B基因表达丰度,差异有统计学意义(P<0.05)。结论:ARID5B基因在卵巢癌组织中呈现低表达,其表达丰度与卵巢癌患者预后呈负相关,该基因可能是血根碱干预卵巢癌细胞的一个靶点。 Objective: To investigate the expression and clinical significance of ARID5B gene in ovarian cancer by collecting relevant data in Oncomine database,and explore the effect of sanguinarine on ARID5B gene in ovarian cancer cells.Methods: The information and expression of ARID5B in the Oncomine database was collected and performed.Kaplan-Meier method was used to analyze the relationship between ARID5B and overall survival time of ovarian cancer patients,and to explore its clinical significance.The ovarian cancer SKOV3 cells were divid- ed into control group and sanguinarine group.Total RNA was extracted after 48 hours.The gene expression profile was detected by whole-genome chip,and the differential genes was screened to investigate the effect of sangui- narine.Results: There were 13 studies referred to ARID5B gene levels in the ovarian cancer and normal ovarian tissues collected in the Oncomine database.The expression of ARID5B gene in ovarian cancer tissues was signifi- cantly lower than that in the normal ovarian tissues( P<0. 05).Survival analysis showed that the disease-free survival and overall survival of ovarian cancer patients with high expression of ARID5B gene were significantly better than those with low expression,and the prognosis of patients with high expression was better( P<0. 05).The san- guinarine significantly up-regulate the abundance of ARID5B gene expression in ovarian cancer SKOV3 cells,and the difference is statistically significant( P<0. 05).Conclusions: There exists lower expression of ARID5B in the tissues of ovarian cancer,and its expression abundance is positively correlated with the prognosis of ovarian cancer patients.This gene may be a new target for sanguinarine to interfere with ovarian cancer cells.
