肾衰Ⅱ号方对慢性肾衰竭大鼠肾组织自噬相关蛋白表达的影响 被引量:19

Effects of Shenshuai Recipe Ⅱ on Autophagy Related Proteins Expression in Chronic Renal Failure Rats′ Nephridial Tissue

摘要 目的肾衰Ⅱ号方对5/6(A/I)肾切除慢性肾衰竭(CRF)大鼠肾组织自噬相关蛋白表达的影响。方法随机选取65只SD雄性大鼠采用5/6(A/I)肾切除术制备CRF大鼠模型,将45只成功造模大鼠随机分为模型组、中药组(肾衰Ⅱ号方10 mL/kg)、西药组(氯沙坦钾6 mL/kg),每组15只,另取15只大鼠作为假手术组。给予相应干预,60日后检测大鼠的肾功能[血肌酐(SCr)、血尿素氮(BUN)、内生肌酐清除率(CCr)]及血红蛋白(HGB),采用Western Blot检测自噬相关蛋白Atg5、Beclin-1、LC3的表达,HE和Masson染色观察肾组织病理形态。结果与本组干预前比较,中药组、西药组干预后SCr、BUN、HGB水平均降低(P<0.01),CCr水平升高(P<0.01)。与假手术组比较,模型组BUN及SCr升高(P<0.01),CCR及HGB降低(P<0.01),肾皮质和肾髓质Atg5、Beclin-1蛋白表达及LC3-Ⅱ/LC3-Ⅰ升高(P<0.01,P<0.05)。与模型组同期比较,两个给药组干预后BUN水平降低(P<0.01), CCr水平升高(P<0.01),肾皮质和肾髓质Atg5、Beclin-1蛋白表达及LC3-Ⅱ/LC3-Ⅰ升高(P<0.01,P<0.05)。HE和Masson染色显示,中药组和西药组肾脏病理变化明显减轻,优于模型组。结论肾衰Ⅱ号方能够改善5/6(A/I)肾切除CRF大鼠的肾功能,提高HGB水平,减轻肾间质纤维化,其作用机制可能与自噬相关信号通路的激活有关。 Objective To observe the effects of Shenshuai RecipeⅡ(SSR) on the autophagy related proteins expression in the chronic renal failure(CRF) rats induced by 5/6 ablation/infarction(A/I). Methods Totally 65 SD male rats were randomly selected to induce CRF rat model by 5/6(A/I) and 45 successful rat models were randomized into model group, Chinese medicine group(SSR 10 mL/kg) and Western medicine group(Losartan potassium 6 mL/kg), 15 rats in each group. Another 15 rats were recruited as sham operation group. After 60-day-intervention, renal function[serum creatinine(SCr), blood urea nitrogen(BUN), creatinine clearance rate(CCr)] and hemoglobin(HGB) were measured, the autophagy related proteins expression of Atg5,Beclin-1,LC3 were determined by Western Blot,the renal pathological changes were observed by HE and Masson staining. Results Compared with the pre-intervention, the levels of SCr,BUN and HGB decreased and the level of CCr increased in the Chinese medicine group and the Western medicine group after the intervention(P<0.01). Compared with the sham operation group, the level of BUN and SCr were increased(P<0.01),the level of CCR and HGB were decreased(P<0.01), the expression of Atg5,Beclin-1 protein and LC3-Ⅱ/LC3-Ⅰ in the renal cortex and the medulla increased in the model group(P<0.01, P<0.05). After intervention, the level of BUN decreased(P<0.01), the level of CCr increased(P<0.01),the expression of Atg5,Beclin-1 protein and LC3-Ⅱ/LC3-Ⅰ in the renal cortex and the medulla increased in medication groups as compared with that in the model group(P<0.01, P<0.05). HE and Masson staining showed that the pathological changes in the Chinese medicine group and Western medicine group were significantly ameliorated, better than those of the model group. Conclusion SSR could improve renal function and relieve renal interstitial fibrosis in CRF rats induced by 5/6(A/I), which could be associated with activation of autophagy related signaling pathway.
