雄激素对再生障碍性贫血患者端粒酶活性的影响 被引量:3

Effect of androgen on telomerase activity in aplastic anemia patients

摘要 目的探讨雄激素对再生障碍性贫血(AA)患者端粒酶活性的影响及端粒在AA发病机制中的作用。方法纳入2016年1月~2017年3月于我院就诊的初诊为AA的患者30例作为试验组,采用随机分组方法将其分为环孢素A(CsA)+雄激素组和CsA组,每组各15例。采用酶联免疫吸附试验(EUSA)双抗体夹心法检测患者治疗前后端粒酶活性。选取同期于我院体检的15例健康者作为对照组,检测其端粒酶活性并与试验组进行比较。结果试验组患者基线端粒酶活性(OD,0.30±0.16)明显高于对照组(0.11±0.06,P<0.001)。CsA+雄激素组端粒酶活性(OD值)在治疗后第3个月、第6个月逐渐降低,而第9个月、第12个月明显升高,治疗前后比较差异有统计学意义(P<0.05),但与CsA组比较差异无统计学意义(P<0.05)。CsA+雄激素组血液学改善率优于CsA组,两组比较差异有统计学意义(P=0.03)。结论雄激素可调节AA患者端粒酶活性,雄激素治疗后端粒酶活性升高以修复损耗的端粒,从而使骨髓造血功能得到恢复。 Objective To explore the effect of androgen on telomerase activity in aplastic anemia(AA)patients and the effect of telomeres in the pathogenesis of AA.Methods A total of 30 patients who came to our hospital and the primary diagnosis was AA from January 2016 to March 2017 were enrolled as treatment group.The treatment group was divided into cyclosporine A(CsA)+androgen group and CsA group randomly with 15 cases in each group.Telomerase activity of patients before and after the treatment was detected by enzyme linked immunosorbent assay(ELISA).The telomerase activity of 15 healthy patients in our hospital was detected and compared with the treatment group.Results Baseline telomerase activity(OD value)in the treatment group(0.30±0.16)was significantly higher than that in the control group(0.11±0.06,P<0.001).In CsA+androgen group,telomerase activity(OD value)decreased at 3 and 6 months after the treatment,but increased at 9 and 12 months.There was significant difference in the group before and after the treatment(P<0.05),but there was no significant difference between the 2 groups(P>0.05).The hematological response of CsA+androgen group was better than that in CsA group(P=0.03).Conclusion Androgen can regulate telomerase activity of AA patients.The telomerase activity increased after androgen therapy and that would repair the lost telomere length,thus the hematopoietic function of the bone marrow was restored.
