黄芩苷通过激活Notch信号通路抑制银屑病角质形成细胞的活力 被引量:26

Baicalin inhibits the activity of keratinocytes in psoriasis by activating Notch signaling pathway

摘要 目的研究黄芩苷对银屑病角质形成细胞活力、细胞周期的影响,并探讨其可能的作用机制。方法噻唑蓝(MTT)实验检测(0、10、50、100、200、300)μg/mL黄芩苷作用于角质形成细胞48h后细胞的活力;流式细胞术检测细胞的周期分布和凋亡率,实时定量PCR检测细胞KI抗原67(ki67)、Fas、胱天蛋白酶3(caspase-3)的mRNA水平;Western blot法检测Notch 1、Jagged 1和发状分裂相关增强子1(Hes 1)蛋白水平。结果黄芩苷抑制角质形成细胞的活力,呈剂量依赖性;200μg/mL黄芩苷显著促进角质形成细胞凋亡,阻滞于S期,抑制ki67水平,增加Fas和caspase-3水平,下调Jagged 1蛋白含量,上调Notch1、 Hes 1蛋白水平。结论黄芩苷能显著抑制角质形成细胞的活力,促进凋亡,可能是通过激活Notch信号通路发挥作用。 Objective To investigate the effect of baicalin on the viability and cell cycle of psoriatic keratinocytes and its possible mechanism. Methods MTT assay was used to detect the viability of keratinocytes treated by 0, 10, 50, 100, 200, 300 μg/mL baicalin for 48 hours. The cell cycle and apoptotic rate were detected by flow cytometry. The mRNA expression levels of ki67, Fas, and caspase-3 were analyzed by real-time quantitative PCR and the protein expression of Notch 1, Jagged 1 and Hes l in the keratinocytes were observed by Western blot analysis. Results The viability of keratinocytes was inhibited by baicalin in a dose-dependent manner. Baicalin(200 μg/mL) significantly promoted the apoptosis of keratinocytes, arrested the S phase, inhibited the mRNA expression of ki67, increased the Fas and caspase-3 levels, down-regulated the protein expression of Jagged 1, and up-regulated the Notch 1 and Hes protein levels. Conclusion Baicalin can significantly inhibit the viability of keratinocytes and promote cell apoptosis, probably by activating Notch signaling pathway.
