摘要
目的:研究四逆散对非酒精性脂肪性肝炎(NASH)大鼠的肝保护作用。方法:采用高脂饮食喂饲12周制备NASH大鼠模型。自第9周起,给予四逆散灌胃治疗共4周。检测血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL)、高密度脂蛋白胆固醇(HDL)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、葡萄糖(GLU)和空腹胰岛素(INS)等指标。测量肝质量、计算肝指数,采用HE染色观察肝组织形态学变化。结果:高脂饲料喂养12周,大鼠血清TC、LDL和GLU水平、ALT和AST的活性及胰岛素抵抗指数(IR)均有升高,肝指数增高,肝细胞肿胀、胞浆内有大小不等、数量不一的的脂滴空泡,汇管区有炎细胞浸润。经四逆散治疗后,明显降低了血清TC、LDL、GLU、IR水平和ALT和AST活性,肝指数明显降低,肝组织内炎症和脂肪变均有明显的改善。结论:四逆散可通过纠正脂代谢紊乱、改善胰岛素抵抗、保护肝细胞等途径,对NASH发挥治疗作用。
Objective: To study the hepatoprotective effect of Sini San on nonalcoholic steatohepatitis (NASH) rats. Methods: NASH rats were fed with high fat diet for 12 weeks. From the 9th week, Sini San was given orally for 4 weeks. Serum total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL), high density lipoprotein cholesterol (HDL), alanine aminotransferase (ALT), aspartate aminotransferase (AST), glucose (GLU) and fasting insulin (INS) were detected. The liver mass was measured and liver index was calculated. The hepatic morphological changes were observed by HE staining. Results: After 12 weeks of high fat diet feeding, the levels of serum TC, LDL and GLU, the activities of ALT and AST, and insulin resistance (IR) index were all increased. The liver index was increased. The hepatocytes swelled, there were fat droplet vacuoles of different sizes and numbers in the cytoplasm, and inflammatory cells infiltrated the portal area. After treatment with Sini San , the levels of TC, LDL, GLU, IR and ALT and AST activity in serum were significantly decreased, the liver index was significantly decreased, and the inflammation and steatosis in liver tissue were significantly improved. Conclusion: Sini San can treat NASH by correcting lipid metabolism disorder, improving insulin resistance and protecting hepatocytes.
