摘要
目的:阐明不同物候期栽培远志药材中次级代谢物的动态变化规律。方法:采用UPLC/Q-TOF MS代谢组学技术,对不同物候期(4月龄至16月龄,10批样本)栽培远志药材中的次级代谢物的含量进行分析。结果:10批栽培远志药材可分为Ⅰ(休眠期)、Ⅱ(花果期)、Ⅲ(休眠期)三个阶段,在推断指认出的31个次级代谢物中,6个蔗糖酯类物质(No.1~6)、14个低聚糖酯类物质(No.10~23)、8个皂苷类物质(No.24~31),在Ⅰ阶段时含量均较低;在Ⅱ阶段前期含量逐渐升高,后期则含量转低,但整体含量均高于Ⅰ阶段;在Ⅲ阶段时,蔗糖酯类物质的含量逐渐升高,而低聚糖酯类、皂苷类物质的含量则有降低趋势。3个■酮类物质(No.7~9)在Ⅰ阶段时含量较高,Ⅱ、Ⅲ阶段与Ⅰ阶段相比含量转低。结论:10批栽培远志药材中,四类次级代谢物的含量会随物候期的不同而呈动态变化,该结果不仅为研究整个物候期栽培远志药材中次级代谢物的动态变化规律奠定基础,并进而为深入研究远志药材中的某些活性成分提供明确的样品采集时间。
Objective:To study the dynamic variation rules of the secondary metabolites in the cultivated Polygala tenuifolia of different phenological phases.Methods:The contents of secondary metabolites in the cultivated Polygala tenuifolia with different phenological phases(10 samples of cultivated Polygala tenuifolia from 4 months to 16 months)were determined and analyzed by UPLC/Q-TOF MS metabolomic technique.Results:Ten samples of the cultivated Polygala tenuifolia were divided into three stages:Ⅰ(dormant stage),Ⅱ(flowering stage),Ⅲ(dormant stage).Among the 31 secondary metabolites were identified by inference,the contents of six sucrose esters(No.1~6),fourteen oligosaccharide esters(No.10~23),and eight saponins(No.24~31)were low in the stage Ⅰ.In the stageⅡ,the contents of above three kinds of compounds increased and then decreased,but compared with stageⅠ,the contents of these compounds were higher in the stageⅡ in general.In the stageⅢ,the contents of sucrose esters increased gradually,while the contents of oligosaccharide esters and saponins tended to go down.Compared with stages Ⅱ and Ⅲ,the contents of xanthone(No.7~9)were higher in the stageⅠ.Conclusion:The contents of four kinds of secondary metabolites in the 10 samples of cultivated Polygala tenuifolia show the dynamic changes with the variation of phenological phases.It′s not only laid a scientific foundation for exploring the dynamic changes of the contents of secondary metabolites in the cultivated Polygala tenuifolia of whole phenological phases,but also provide a definite sample collection time for further study of some active components of cultivated Polygala tenuifolia.
