摘要
细胞生长依赖细胞微环境,细胞外基质是细胞微环境的重要组分,不仅为细胞提供了支持、居住环境,同时通过膜受体而影响细胞的黏附、迁移、增殖、分化、存活等生物行为。目前广泛应用的体外二维细胞培养与细胞的体内生长状态差别较大,三维细胞培养是利用三维结构的细胞外基质模拟体内环境,使细胞在近似体内环境中生长。三维培养中涉及诸多影响因素,如细胞外基质的组成、支架结构、硬度及微流控芯片调控营养物质供应及代谢。本文对国内外该领域的研究进行综述整理并提出展望。
Cell growth depends on the microenvironment.The extracellular matrix is an important component of cell microenvironment,which not only provides support and living environment for cells,but also affects the biological behaviors such as cell adhesion,migration,proliferation,differentiation and survival of cells through membrane receptors.Currently widely used in vitro culture system is two-dimeosional culture,which do not simulate the living environment in vivo,while three-dimensional cell culture can simulate the body environment very well and make cells approximately reflect the growth situation in vivo.There are many influencing factors in three-dimensional culture,such as the composition of extracellular matrix,scaffold structure,hardness and microfluidic chip to regulate nutrient supply and metabolism.This article reviews the research in this field at home and abroad and puts forward prospect.
